Objective: To understand the function of TRPV1 in endothelial cell apoptosis induced by hyperhomocysteine.and the potential mechanism of capsaicin attenuates atherosclerosis.Methods:To cultivate human umbilical vein endothelial cells use different concentration and different time of homocysteine,CCK-8 was used to evaluate the proliferation of HUVECs.TUNEL staining was used to evaluate the apoptosis.Western Blotting is using to assess the expression of TRPV1 protein.Results:(1)The proliferation of HUVECs were significantly inhibited by homocysteine in a dose-dependent manner and the OD value was declined from(1.03±0.12)to(0.22±0.01)(F=11.73,P=0.000),homocysteine intervention at0.5mmol/L for 24 hours has inhibiting effect on proliferation of human umbilical vein endothelial cells(P<0.05);(2)There were TRPV1 expression on human umbilical vein endothelial cells;(3))The expression of TRPV1 and cell apoptosis were significantly different compared with control group(P<0.05);(4)Compared with homocysteine group,after intervention using capsaisin,the expression of TRPV1 protein was increased(P<0.05)and the apoptosis decreased(P<0.05)Conclusion:Hyperhomocysteine can downregulate the expression of TRPV1 protein and promote the apoptosis of endothelial cells.Capsaisin can inhibit the apoptosis of endothelial cells induced by hyperhomocysteine.
