Objective Effects of shi-da-la-zhi-wan on Notch1 receptor expression in hippocampus of rats with focal cerebral ischemia-reperfusion,to explore the mechanism of repair and protection of ischemic lesions and neurons.Methods The 102 SD rats were randomly divided into 17 groups of 6,namely:control group,sham operation group,and the model group,nimodipine group,shi-da-la-zhi-wan low dose group,shi-da-la-zhi-wan middle dose group and shi-da-la-zhi-wan high dose group1 d,3d,7d group.In addition to blank group,sham operation group,The other groups were treated with suture method for focal cerebral ischemia and reperfusion.Wherein the control group,sham operation group,model group were fed with normal saline.nimodipine group was given a concentration of 1.08g/L gavage.shi-da-la-zhi-wan group respectively with a concentrations of 4.5g/ml,3.0g/ml and 1.5g/ml gavage.over the same time of day each group orally,twice a day.Respectively in the 1d,3d,7d collect the brain tissue.The degree of recovery of nerve function defect score using Garcia JH.Immunofluorescence,Western-blot,RT-PCR detection of ischemic brain tissue expression of Notch1.Results1.Nerve function score results(1)There was no significant difference between the blank group and the sham operation group at each time point(P>0.05).(2)There was significant difference between the model group and the drug group at each time point compared with the blank group and the shamoperation group(P<0.05).(3)The time points of each drug group were statistically significant compared with the model group(P<0.05).(4)There was significant difference between the nimodipine group and the shi-da-la-zhi-wan low dose group at each time point(P<0.05),compared with the shi-da-la-zhi-wan mid-dose group at each time point was not statistically significant(P>0.05),compared with the shi-da-la-zhi-wan high dose group,the difference was significant at each time point(P<0.01).(5)Shi-da-la-zhi-wan low dose group compared with shi-da-la-zhi-wan middle dose group and shi-da-la-zhi-wan high dose group at each time point was statistically significant(P<0.05 or P<0.01).2.Notch1 immunofluorescence middle dose group and shi-da-la-zhi-wan high dose group at each time point was statistically significant(P<0.05 or P<0.01).Conclusions1.Shi-da-la-zhi-wan on cerebral ischemia and reperfusion injury neurological deficits have significant improvement.2.Effect of shi-da-la-zhi-wan on cerebral ischemia reperfusion injury,may be related to activation of Notch signaling pathway plays Notch1 receptor or promote repair of ischemic neurons,thereby improving impaired brain function related.
