Objective: To study the expression of cIAP protein and Caspase-7 protein in different endometrial tissues,the relationship between the expression of these two genes and the clinical features of endometrial adenocarcinoma(FIGO 2009),pathological grade and lymph node metastasis were analyzed,and the correlation between the expression of the two,objective to investigate the significance of the two expression in the endometrium.Methods: Selected 84 cases of endometrial specimens from pathology department of the First Affiliated Hospital of Henan University of Science and Technology from 2008 to 2010.According to the different histological types,they were divided into 3 groups: normal proliferative endometrium group(n=12),endometrial atypical hyperplasia group(n=17)and endometrial adenocarcinoma group(n=55).The expression of cIAP protein and Caspase-7 protein in each group were analyzed by immunohistochemical SP technique.The relationship between cIAP protein and Caspase-7 protein and the clinical features of 55 patients with endometrial adenocarcinoma were analyzed,including clinical stage,pathological grade and myometrial invasion.55 cases of endometrial adenocarcinoma patients underwent surgical staging,including: I~II stage in 37 cases,III~IV stage in 18 cases,a total of 45 patients with lymph node dissection,lymph node metastasis was found in 18 cases.The correlation between the expression of cIAP protein and Caspase-7 protein in 55 cases of endometrial adenocarcinoma was analyzed.The selected tissue samples of patients had no other malignant tumors,the patients before the operation were treated without radiation and chemotherapy,all the specimens were fixed by the department of Pathology of the First Affiliated Hospital of Henan University of Science and Technology,by two and above senior physician readers.The experimental data using SPSS17.0 statistical software,using ?2 test,Spearman correlation analysis,using P<0.05 as the difference was statistically significant.Results: The positive signal of cIAP protein and Caspase-7 protein in endometrial epithelial cells localized in the cytoplasm,brownish yellow granules,the positive expression of cIAP protein were: endometrial adenocarcinoma group(43/55,78.2%),endometrial atypical hyperplasia group(5/17,29.4%),normal endometrium group(3/12,25.0%),with significant differences between the three groups(P<0.05).The expression of cIAP in endometrial adenocarcinoma associated with clinical stage,pathologic stage,lymph node metastases(P<0.05),and has nothing to do with the muscular layer infiltration and patient's age(P>0.05).The positive expression of Caspase-7 protein were: endometrial adenocarcinoma group(20/55,36.3%),atypical endometrial hyperplasia group(13/17,76.4%),normal endometrium group(10/12,83.3%),there was significant difference between three groups(P<0.05).The expression of Caspase-7 in endometrial adenocarcinoma associated with clinical stage,pathologic stage,lymph node metastases(P<0.05),and has nothing to do with the muscular layer infiltration and patient's age(P>0.05).Spearman correlation analysis showed that there was a negative correlation between cIAP protein and Caspase-7 protein in 55 cases of endometrial adenocarcinoma(rs=-0.607,P<0.05).Conclusions: 1.Compared with normal endometrium group and atypical endometrial hyperplasia group,the apoptosis inhibitory protein cIAP was highly expressed in the endometrial adenocarcinoma group,and the expression of apoptosis protein Caspase-7 was low in the endometrial adenocarcinoma group,suggesting that the disorder of apoptosis may be involved in the malignant transformation of endometrial tissue.2.The expression of cIAP and Caspase-7 in endometrial adenocarcinoma was correlated with pathological grade,clinical stage and lymph node metastasis,and has nothing to do with the patient's age and myometrial invasion,suggesting that the two may play a role in the progression of endometrial adenocarcinoma invasion.3.cIAP was negatively correlated with the expression of Caspase-7 in endometrial adenocarcinoma tissues,suggesting that the two mutually restricts in the development of endometrial adenocarcinoma,two factor combined detection may provide a theoretical basis for evaluating the clinical prognosis of endometrial adenocarcinoma.
