| Objective To research the effect of lycium barbarum polysaccharide(LBP)on oxidative damage and P38 MAPK,caspase-8,caspase-3 and Bcl-2 content of HaCaT cells irradiated by UVB.Methods LBP powder was obtained by ultrasonic assisted water extraction.HaCaT cells were cultured in vitro and they were randomly divided into control group,UVB irradiation group(30mJ/cm2 radiation 40 min UVB)and UVB+LBP group(UVB 30mJ/cm2 radiation 40 min + LBP 1mg/ml).Twelve hours before UVB irradiation,LBP was added into the culture medium in the UVB+LBP group,and after irradiation,the cells were continuously cultured in the incubator of 20 hours.The morphological changes and apoptosis of the cells were observed by inverted microscope;MTS assay was used to detect the A value of each group;SOD,CAT,GSH-Px and MDA content of the cells in each group were tested by enzyme linked immunosorbent assay;The contents of P38 MAPK,caspase-8,caspase-3 and Bcl-2 in each group were detected with Western blot.Results Compared with the control group,in UVB irradiation group,he morphological of HaCaT cells markedly changed,the number of floating dead cells increased significantly and the A value decreased significantly(P<0.05);Compared with UVB irradiation group,in UVB+LBP group,the morphology of HaCaT cells tented to be normal,the number of floating dead cells decreased significantly and the A value increased significantly(P<0.05).Compared with the control group,SOD activity,CAT activity and GSH-Px content of the UVB irradiation group were significantly decreased,and the content of MDA increased significantly(P < 0.05);Compared with UVB irradiation group,SOD activity,CAT activity and GSH-Px content in UVB+LBP group increased significantly,and MDA content decreased significantly(P < 0.05).Compared with the control group,P38 MAPK,caspase-8,caspase-3 content of the UVB irradiation group increased significantly,and the content of Bcl-2 decreased significantly(P<0.05);Compared with UVB irradiation group,P38 MAPK,Caspase-8,caspase-3 content of UVB+LBP group decreased significantly,Bcl-2 content increased significantly(P<0.05).Conclusion LBP could significantly alleviate the oxidative damage,down-regulation content of P38 MAPK,caspase-8 and caspase-3 and up-regulation content of Bcl-2 caused by UVB irradiation of HaCaT cells. |
