Objective: To investigate the expression and function of IL-6 and TGF-?1 in urethral injury.Methods:1 A model of urethral injury was established by traumatic injury of mouse urethral tissue and the morphological changes were observed and evaluated by hematoxylin-eosin staining(HE)in normal urethra and urethral injury mice.2 Expression of IL-6 and TGF-?1 mRNA in normal urethra and urethral injury mice was determined by q RT-PCR.3 In situ hybridization combined with MAC2 was performed to locate the expression of IL-6,TGF-?1 gene.Results:1 The hyperplasia and thickness of the mucosa layer in urethral t issue increased significantly.Morphometric analysis showed that the urethral tissue hyperplas ia of the urethral injury tissues was significantly increased.The thickness of the mucosa layer(303.2 ± 21.4?m)in urethral injury group was significantly increased(P <0.05)compared with that in normal urethra group.The rate of urethral stenosis was 90.67%,which was significantly higher than that of normal urethral tissue(41.56%,P <0.01).2 The expression of IL-6 and TGF-?1 mRNA in urethral injury was significantly higher than that in normal urethral tissue.q RT-PCR analys is revealed that IL-6 and TGF-?1 mRNA expression was elevated by only 0.9-fold and 1.2-fold in normal urethra,while 2.5-fold and 4.3-fold in urethral injury respectively.3 Inflammatory infiltration in urethral injury increased,mainly due to increased expression of inflammatory factors IL-6,TGF-?1 and macrophage infiltration.In situ hybridization combined with MAC2 immunostaining suggested that IL-6 probe-MAC2 and TGF-?1 probe-MAC2 positive rate were significantly higher in urethral injury than normal urethral tissue.Conclusions:High expression level of IL-6 and TGF-?1 promotes the activation and infiltration of inflammatory cells.They interact with each other,leading to urethral mucosal tissue chronic hyperplasia,fibrosis,promoting the formation and development of urethral stricture.
