Objectives: To investigate the relationship between the expression of Th22 cells and its functional factors IL-22 and clinical stage and Ca153 in the peripheral blood of patients with breast cancer by detecting the levels of Th22 cells,cytokines IL-22 and cancer antigen Ca153.Methods:1 Object screeningFrom January 2015 February-2016 year in medical oncology,Affiliated Hospital of Hebei University who has breast surgery and pathological diagnosis of 40 cases of breast cancer patients as a group of breast cancer,40 cases of healthy persons from the medical department of the Affiliated Hospital of Hebei University as the control group.There were no other serious diseases in the breast cancer group.2 Plasma samples were collectedSerum samples were collected from two groups of patients in the early morning,fasting peripheral venous blood 10 ml,centrifugal supernatant,make the mark.3 Effect factor detectionELISA method to detect the concentration of cytokine IL-22 in peripheral venous blood.4 Th22 cell detectionFlow cytometry was used to detect the frequency of Th22 cell /CD4+T cell expression in peripheral venous blood.Results:1 The frequency of Th22 cell /CD4+T in peripheral blood of breast cancer patients was significantly higher than that in healthy controls(P <0.05)2 The plasma IL-22 levels in breast cancer patients in experimental group was 24.32 pg/ml higher than the control group of healthy people the level of plasma IL-22 in 12.98 pg/ml(P < 0.01);there is a significant positive correlation between plasma IL-22 level and clinical stage of breast cancer between(r= 0.782,P < 0.05),and increased with increasing of clinical stages;there is a positive correlation between the size and lymph node in peripheral blood plasma IL-22 levels of cytokines and tumor metastasis in patients with breast cancer(r=0.234;r=0.196,P < 0.05).3 Ca153 group of breast cancer patients with experimental average level was 49.32 U/ml,significantly higher than the control group of 11.31 U/ml(P< 0.01);there is a significant positive correlation between plasma Ca153 level and clinical stage of breast cancer between(r= 0.793,P < 0.05),and increased with the increasing level of clinical staging increased;there is a positive correlation between the size of lymph node and peripheral blood plasma levels of Ca153 in patients with breast cancer metastasis(r=0.178;r=0.206,P< 0.05),were highly expressed in lymph node metastasis group.4 The level of IL-22 and Th22 cells showed significant positive correlation between the frequency(r=0.826,P < 0.05);the level of Ca153 and Th22 cells showed a significant positive correlation between the expression of frequency(r=0.893,P < 0.05),the level of Ca153 and IL-22 cytokine levels showed significant positive correlation(r=0.845,P < 0.05).Conclusions:1 Th22 cells and IL-22 were significantly higher in breast cancer patients,the two were positively correlated,suggesting that Th22 cells and their cytokines IL-22 are closely related to the occurrence of breast cancer;2 IL-22 was positively correlated with tumor size,lymph node metastasis and clinical stage,suggesting that IL-22 may promote the growth and metastasis of breast cancer cells,which is closely related to its development;3 Th22 cells,IL-22 cell factor level and the expression level of Ca153 was positively related to the combined detection of expression level and CA153 Th22 cells and IL-22 can provide better theoretical basis at thecellular level and plasma level of recurrence or progression. |
