Background: During assisted reproduction technology(ART),Assessing embro quality,selecting embryo for translation is critical for pregnancy sussess in IVF-ET treatment.At present,the widespread adoption of the single morphological evaluation method to select embryos,but this kind of method to predict the embryo development and implantation of potential ability is limited,need to find out the better noninvasive predictor of embryo implantation potential.In recent years many studies have shown that embryo culture human chorionic gonadotropin(beta hCG)and interleukin 1(IL-1 beta)have correlation with IVF embryos development potential,are expected to become noninvasive indexes of clinical transplantation embryos,but there are still controversy on account of the different culture conditions and the different detection assays in laboratories.Objective: To study the validation of beta hCG and IL-1 beta can be as IVF embryo development and implantation potential markers,as IVF-ET embryo transfer provide noninvasive selection criteria,further improve the clinical pregnancy rate,reduce the number of transferred embryos,reduce the occurrence of multiple pregnancies,thereby improve IVF outcome.Methods:1 Object of study Choose In December 2016 was conducted in the second hospital,hebei medical university reproductive medicine in vitro fertilization and embryo transplantation(In vitro fetilization-embryo transfer,IVF-ET)to help patients with pregnancy embryo transplantation,the number of transferred embryos is two,collect two cultures of transferred embryos and the droplets that do not cultivate embryo as blank controls.2 Test method Beta hCG levels in embryo culture medium was detected by electro chemiluminescence immuno-assay(ECLI),using enzyme-linked immunosorbent assay(ELISA)to detect the levels of IL-1 beta in embryo cultures.Collect clinical and embryology laboratory data.3 Grouping According to IR(0%,50%,100%)in three groups comparison between groups of clinical and laboratory data and the levels of beta hCG and IL-1 beta.According to the hCG dose group comparison beta hCG levels.Grouped by IR respectively to compare the blood P secretion and the levels of IL-1 beta.Results: 1 Embryonic beta-hCG horizontal comparison 1.1The average concentrations of beta-hCG in 0%IR,50%IR and 100%IR were 0.79±0.47,1.49±0.61,1.70±0.53 IU/L,respectively,comparing the three groups had statistically significant difference(P<0.05).Morpholo-gical grading in the three groups were13.34±2.70,15.50±1.48,15.30±1.05,respectively,comparing the three groups had statistically significant difference(P<0.05).1.2According to the age of grouping,?35 years old group beta-hCG levels was 1.11 ±0.55 IU/L,>35years old group beta-hCG levels was 1.09 ±0.95 IU/L,comparing the two groups had no statistical difference(P>0.05).According to fertillization methods of grouping,beta-hCG levels of IVF group was 1.07 ±0.67 IU/L,beta-hCG levels of ICSI group was 1.20 ±0.62,compa-ring the two groups had no statistical difference(P>0.05).According to the hCG dose of grouping,in the ?6500IU group,beta-hCG levels was 1.10 ±0.65 IU/L;in the >6500IU group,beta-hCG levels was 1.16 ±0.60 IU/L.comparing the two groups had no statistical difference(P>0.05).2 Embryonic IL-1 beta horizontal comparison 2.1 The average concentrations of IL-1beta in 0%IR,50%IR and 100%IR were 0.27±0.24,1.07±2.91,8.39±16.89 IU/L,respectively,comparing the three groups had statistically significant difference(P<0.05).Morphological grading in the three groups were 13.08±2.06,15.17±1.53,13.88±1.64,respec-tively,comparing the three groups had statistically significant difference(P< 0.05).2.2 According to the age of grouping,?35 years old group IL-1beta levels was 1.62±6.37 IU/L,>35years old group IL-1beta levels was 0.44±0.57 IU/L,comparing the two groups had no statistical difference(P>0.05).According to fertillization methods of grouping,IL-1beta levels of IVF group was 1.68 ±6.75 IU/L,IL-1beta levels of ICSI group was 0.59±0.80 IU/L,comparing the two groups had no statistical difference(P>0.05).Grouped by IR respectively comparing blood P and the levels of IL-1beta,the results was with no correlation(P>0.05).Conclusions: Beta-hCG and IL-1beta in embryo cultures by embryos secretion may be important indexes of embryo development and implantation potential,they will provide non-invasive indicators for selecting of translated embryos.
