Objective: In our country,Esophageal carcinoma is a kind of the characteristics tumors,and Hebei province have a highest incidence of esophageal Esophageal carcinoma.In the clinic screening,the results shows that most patients were missed the best treatment stage.Although manay researchs and methods of the esophageal carcinoma treatment have been reported,but there is a subtypes of esophageal carcinoma which was rare and high malignant degree: basaloid squamous cell carcinoma of the esophagus(BSCCE)is limited by its rarity in the study.As the reports,the best treatment of BSCCE is surgical,However,its Overall survival is far from ideal,For the further study,with the pathological characteristic of BSCCE and excavate potential therapeutic targets,so as to improve the Overall survival for the clinical treatment,This study using bioinformatics methods and the technology of gene chip,The tissue samples of BSCCE were analyzed,The expression gene which related to the possible process of pathology and tumor immune escape mechanism will be discuss and assay,At the same time,the Overall survival and tumor microenvironment of BSCCE has be discussion,in order to provide a better treatment in clinical medical for BSCCE.Methods: This experiment using bioinformatics method,the gene chip of group gained from clinical samples by using R language software(R 3.2.3)were obtained the original data,For the complete the quality control,the data normalization and noise reduction processing have be finished,Independent access to reliable BSCCE tissue samples and parts of gene differential expression in normal tissue samples(log | FC| ?2,P <0.01),DAVID online analysis system was used to analyse for the differentially expressed genes of gene ontology(GO)and kyoko encyclopedia of genes and genomes(KEGG)for the signaling pathway analysis and the analysis of the differences in gene Disease(diseases)to get the analysis results of histo-pathological situationrelated BSCCE,using STRING-OL tools for the credibility of the differentially expressed genes(credibility 0.4 or higher)screening,trying to reduce false positives may get Protein interaction analysis of differentially expressed genes(Protein-Protein interaction,PPI),and further the PPI results after using Cytoscape software for screening make the interaction network diagram of corresponding gene,and further screening core of differences gene expression.Using the above analysis further understand the pathological state of BSCCE is neutrophils into reasons for its unique and Overall survival of malignant must be analyzed.Results: the rise in relative to the normal BSCCE tissue express 489up-regulate expressed genes,And 922 down-regulate expressed genes(log |FC |?2,P <0.01).1 GO analysis : In the up-regulate expressed genes of BSCCE tissue,There are 238 GO trems in the results,The GO:0030574-collagen catabolic process,16 genes,GO:0030198-extracellular matrix organization,24 genes,and GO:0005578-proteinaceous extracellular matrix,25 genes which related with Extracellular matrix(ECM),in this research,Such as ITGB4?COL5A1?LAMB3,et al 13 regulate expressed genes were selected..2 KEGG analysis: In the up-regulate expressed genes of BSCCE tissue,Such as p53 signaling pathway,Cytokine-cytokine receptor interaction pathway,Toll-like receptor signaling pathway.In the down-regulate expressed genes,The results that Drug metabolism-cytochrome P450 pathway,MAPK signaling pathway,Calcium signaling pathway,and Cytokine-cytokine receptor interaction pathway.All of these signaling pathway have the correlation with tumor microenvironment.TLR3 which belong to the Toll-like receptor signaling pathway,in this research,Such as LAMB3?LAMC2?MFAP2,et al 11 regulate expressed genes were selected..3 disease analysis: the 605027-Lymphoma,non-Hodgkin,somatic is a important result.RAD54 B and RAD54 L is the point.4 The screening and analysis for the core difference expressed genes: The STRING-OL tool for the screening in PPI analysis,30 core differenceexpressed genes have be selected(confidence?0.4,interaction?20),In the up-regulate expressed genes,there are 24 genes,6 results for the downregulate expressed genes.Such as TOP2A?CDC20?CDC6,et al 15 regulate expressed genes were selected.Conclusion: Some more tumor markers(kinds of 25)could be realized as TGFBI,LAMC2,MFAP2,et al.This research is a base for more BSCCE or SCC clinical study and treatment.
