Parkinson's disease(PD)is a progressive and neurodegenerative disorder with continuous loss of dopaminergic neuronal in the substantia nigra(SN),resulting in the depletion of dopamine in the striatum(ST).It is estimated that there are more than 2% people over 65 years old who suffering from Parkinson diseases.There are 60000 new patients with parkinson's disease each year around the global.It also leads to motor impairment,such as rigidity,bradykinesia,postural instability,and tremor.Recently,DA system has caused more and more attention.Until now,there is no definite conclusion of the pathogenesis of Parkinson's disease.Recently,some researchers found that the high expression of inflammatory factors in Parkinson's disease.Although L-DOPA treatment could alleviate the symptoms of PD for a period of time,long-term administration of L-DOPA always produces dyskinesias.Aucubin is a natural active compound extracted from plants.There are some reports about the active compounds with anti-inflammatory.Morever,our previous study has shown aucubin can protect hippocampal neuronal cells in diabetic rats model,but the mechanism is not very clear.Therefore,the study use MPTP to establish parkinson's disease,and then using aucubin to treat the animals.The study would like to explore whether aucubin have neuroprotective effect on parkinson's disease and its mechanism.It aims to provide a theoretical basis for treating parkinson's disease.Here are the contents and results of this paper.(1)Establishment of MPTP subacute Parkinson model.C57 BL / 6 mice were treated with neurotoxin MPTP for consecutive 7 days.At the same time,treating with different doses of Aucubin for 14 days.The behavioral experiments were carried out on the 1st,3rd,5th,7th and 14 th day.The results showed that aucubin could improve the coordination of PD mice to a certain extent.(2)Neuroprotective effect of aucubin on PD mice.Through HE staining and immunohistochemical staining of the midbrain,we found that Aucubin can inhibit the infiltration of inflammatory cells in the midbrain,and increase the TH-positive cells.In addition,we also found that the production of TH protein in substantia nigra and striatum of PD mice was improved by aucubin.(3)Neuroprotective mechanism of aucubin on PD mice.The NF-kB pathway-associated proteins were detected by Western blot,and then docking acucubin with NF-kB.In conclusion,this study clarifies the neuroprotective effect of aucubin on MPTP-induced PD mice and provides a treatment strategy for the treatment of PD.
