Structure optimization of active natural products is always one of the most important strategies for research and development of innovative drugs,which is the target of natural pharmaceutical chemists to keep fighting.The contents you will find in this paper mainly include some works based on a bioactive natural products,dendroflorin,isolated from Dendrobium densiflorum Lindl..The details were summarized as follows: firstly,total synthesis of dendroflorin was finished for the first time,with a 10-step routine and the overall yield 5.5%;Secondly,26 compounds had been synthesized in two phases based on the structure of dendroflorin for improving the inhibitory activity of PC-3;Thirdly,accidentally,possible cation-? interaction was found during the total synthesis of dendroflorin,and base on that,some preliminary study had been moved forward;Fourthly,design and synthesis of inhibitors against USP-28 with RA1-1,a derivative from dendroflorin,as the leading compound.51 compounds were synthesized in this part.
