Cancer is a serious challenge to the scientists of medical and related fields.The activities of tumor cells are closely related with the bodies' immune system.Therefore,immunotherapy targeted to immune system is a promising research interest for the treatment of cancer.Antitumor vaccines is not only a safer treatment but also aimed at the early stage of the process in which the immune system attack tumor cells.Antitumor vaccine also can avoid the risk of recurrence frequently happened in surgery.Through activating the immune system,antitumor vaccine can also enhance the treatment effect of immune checkpoints blocking and adoptive cell therapy for those lack tumor specific T cells.Above all,to develop highly effective antitumor vaccines is of great importance.MUC1 glycopeptide belong to tumor associated antigen.It has been widely applied in the diagnose and treatment of tumor and have showed good results.Many studies has worked on the MUC1 glycopeptide antitumor vaccine.And considering the weak immunogenicity of glycopeptide antigen,there has been many strategies to overcome it.For example,adding new component into vaccine to construct multiple component antitumor vaccine.Widely used in the vaccine are the carried proteins,Th cell epitope and immunostimulants.However,the effect of antitumor vaccines based on MUC1 glycopeptide antigen are quiet unsatisfactory.Based on the studies before,we applied nanotechnology into the design of MUC1 glycopeptide antitumor vaccine.We introduce the biodegradable polymer,?-PGA,into the vaccine.?-PGA are negative charged and can self-assembly into nanovaccine with the positively charged MUC1 glycopeptide and Th cell epitope.The formation of the vaccine can avoid the difficulty of synthesis and purification in chemical covalent attachment between two or more components.Besides,nanovaccine are more stable and easily carried to the DC cells,which can greatly increase the chance of antigen uptake and process by DC cells.The self-assembly antitumor nanovaccine shows good antitumor results in the evaluation in vitro and in vivo.Self-assembly nanovaccine based on electrostatic interaction can be widely used in the construction of antitumor vaccine in the future.And our work offer a new approach in the development of the MUC1 glycopeptide antitumor vaccine.
