Objective: To investigate the expression of interferon regulatory factor 4 in Jurkat Cells and the effect of adriamycin and bortezomib on interferon regulatory factor 4 and P glycoprotein.Methods: 1)The effects of bortezomib and doxorubicin on the proliferation of Jurkat cells were detected by CCK8 method.2)The effects of doxorubicin and bortezomib and two drugs on the expression of IRF4 m RNA and P-gp m RNA in Jurkat cells at different time were detected by RT-PCR.3)Western Blot was used to detect the effect of doxorubicin and bortezomib and the two drugs on the expression of IRF4 protein in Jurkat cells at different time.Results: 1.Compared with the control group,adriamycin had an inhibitory effect on the proliferation of Jurkat cells.When the concentration of adriamycin was 400 ug / L,the inhibitory effect was the strongest,with the extension of the role of time,the inhibition effect was increased gradually(P <0.05).Bortezomib inhibited the proliferation of Jurkat cells in a time-dependent manner(P <0.05).2.Compared with the control group,the expression of IRF4 m RNA in adriamycin group had no significant difference(P> 0.05).The expression of IRF4 m RNA in the bortezomib group was down-regulated(0.66 ± 0.10)at 48 hours and the most significant effect(0.34 ± 0.09)at 72 hours(P> 0.05).The expression of IRF4 m RNA was down-regulated at 48 hours after the combination of the two drugs.3.There was no significant difference about the expression of P-gp m RNA between adriamycin group and control group(P>0.05).The expression of P-gp m RNA in the bortezomib group was higher than that in the control group(P <0.05).The expression of P-gp m RNA was down-regulated at 48 hours and 72 hours(0.79 ± 0.02,0.59 ± 0.00).After treated with two drugs,the expression of P-gp m RNA was down-regulated(0.73 ± 0.00), which was the most significant in 72 hours(0.47 ± 0.00).4.The expression of IRF4 protein was not significantly affected by adriamycin at each time(P> 0.05).The expression of IRF4 protein was down-regulated at 48 hours and 72 hours in the bortezomib group(P <0.05).The expression of IRF4 protein in combination of two group was down-regulated at 48 hours,which was the most significant in 72 hours(P<0.05).Conclusion: 1?There was positive expression of interferon regulatory factor 4 in Jurkat Cells;2?Adriamycin can inhibit Jurkat cell proliferation,but can not inhibit the expression of interferon regulatory factor 4 and P glycoprotein;3?Bortezomib inhibits the proliferation of Jurkat cells,and its mechanism may be by down-regulating the expression of interferon regulatory factor 4 and P glycoprotein.
