Objective Investigate the effect of metformin on the invasion and migration ability of high metastatic ovarian cancer cell line SKOV3,to study the effect of metformin on the expression of MTA1 in ovarian cancer cell line SKOV3.Methods Highly metastatic ovarian carcinoma cell line SKOV3 was cultured in vitro and the cells were treated with various concentration of metformin.The lateral migration ability was measured by scratch test.Transwell migration experiments were used to detect the ability of cells to penetrate fibrous membranes.Transwell invasion assay were used to detect the invasion and migration ability of cells.The effect of SKOV3 on the ability of adhesion was evaluated by adhersion assay,The expression of MTA1 protein was detected by western blot 72 hours after treatment,and the m RNA expression of MTA1 was evaluated by realtime quantitative PCR.The data was analysis by SPSS17.0,P <0.05 for the difference was statistically significant.Results 1 The ovarian cell line SKOV3 was adherent cell,it grew slowly after metformin was added.There was more particles appeared in the cytoplasm,and the number of cell debris in the culture medium increased after metformin was added.2Compared with control group the lateral migration ability of the experimental group was significantly decreased(P<0.01).With the increase of metformin concentration,the lateral migration of SKOV3 cells was inhibited(P<0.05),With the prolongation of metformin,the lateral migration distance of the cells gradually shortened,when the metformin concentration remained unchanged.3 Transwell migration test showed that the permeability of SKOV3 cells in the experimental group was significantly lower than that in the control group(P<0.01),and the membrane permeability was enhanced with the increase of metformin concentration(P<0.05).4 Compared with control group the cell invasion was in a dose-dependent manner when the SKOV3 cells were treated by metformin for 72h(P<0.01).5 Compared with the control group,the cell adhesion ability of the experimental group was significantly increased in a dose-dependent manner.6Real-time quantitative PCR showed that the m RNA expression level of MTA1 was decreased in metformin treatment groups,significantly different from control group(P<0.05).7 Western blotting showed that the protein level of MTA1 was significantly decreased in metformin treatment groups when compared with control group(P<0.05).Conclusion 1 Metformin inhibited the migration ability of SKOV3 cells,what was correlated with the concentration;2 Metformin inhibited the invasive ability of SKOV3 cells,what was in a dose-dependent manner;3 Treatde by metformin,the expression of MTA1 m RNA in SKOV3 cells was decreased;4 Treatde by metformin,the expression of MTA1 protein in SKOV3 cells was decreased;5 Metformin may inhibit the invasion and migration of SKOV3 cells by inhibiting MTA1 expression.
