Objectives The expression of GDF-15,ZEB1 and TIP30 in gliomas and normal brain tissues were obtained by immunohistochemistry.To explore the possible relationship and possible mechanisms between GDF-15,ZEB1 and TIP30 in the occurrence and development of glioma lesions,and provide a new research basis and theoretical basis for the further study of the pathogenesis,occurrence and development of glioma,and provide more guidance for the clinical treatment of glioma.Methods Select the surgical removal of glioma tissue specimens(all specimens are stored in liquid nitrogen)since 1998 Kailuan general hospital neurosurgical income of 69 patients included in the experimental group,and then 15 cases of normal brain tissue after cerebral hemorrhage resection into the control group.The experimental group was then divided into three groups according to the World Health Organization(WHO)classification and classification of central nervous system tumors(2007 edition):the first group: grade I-II glioma,27 cases;the second group: grade Ⅲ glioma,22 cases;group III:grade Ⅳ glioma,20 cases;the experimental group was divided into two subgroups according to tumor size:more than 5cm group,38 cases;less than 5cm group,31 cases.The gene expression of GDF-15,ZEB1 and TIP30 was obtained by routine HE staining and immunohistochemical staining.Finally,through the SPSS 17.0 Chinese version to carry out statistical analysis,positive rate between groups using chi-square test,the relationship between the positive rate of three factors and the level of glioma was analyzed by spearman rank correlation analysis,comparison of the correlation between the positive expression rates of the three factors was analyzed by spearman rank correlation analysis.Results 1 TIP30 protein in the control group and glioma group were 86.7%(13/15)and47.8%(33/69)respectively.and in I-II subgroup,III subgroup and IV subgroup were 66.7%(18/27),45.5%(10/22),25.0%(5/20),and in the group of less than 5cm and greater than5 cm were 64.5%(20/31)and 34.2%(13/38),the difference was statistically significant(P<0.05).2.The positive rates of GDF-15 protein in the control group and glioma group were20.0%(3/15),55.1%(38/69),the positive expression rates in the I-II,III,IV groups were37.0%(10/27),59.1%(13/22),75%(15/20),the positive rates of expression in the group of more than 5cm group and less than 5cm group were 65.8%(25/38),41.9%(13/31),the difference was statistically significant(P <0.05),3 The positive rates of ZEB1 protein in the control group and glioma group were 6.7%(1/15)and 34.7%(24/69)respectively,the positive expression rates in the I-II,III,IV groups were 14.8%(4/27),40.9%(9/22),55.0%(11/20),the positive rate of expression in the group of less than 5 cm and in the group of greater than 5 cm was 19.4%(6/31),47.4%(18/38)respectively,the difference was statistically significant(P <0.05),4 TIP30 was negatively correlated with GDF-15expression(r =-0.261,P = 0.017),there was a positive correlation between ZEBI and GDF-15 expression in gliomas(r = 0.250,P = 0.022),there was a negative correlation between TIP30 and ZEB1 expression(r =-0.350,P = 0.001).Conclusions 1 The positive rate of expression GDF-15,ZEB1 in the glioma group was significantly increased,in the control group,the positive expression rate was low,suggesting that GDF-15,ZEB1 and glioma are closely related,the positive expression rate of GDF-15 and ZEB1 increased with the increase of malignant grade of glioma,suggesting that GDF-15,ZEB1 and glioma malignant level was positively correlated;2 TIP30 expression in the glioma group was lower than that in the control group,suggesting that TIP30 and glioma are closely related,TIP30 decreased with the increase of the malignancy of glioma in each subgroup,suggesting that TIP30 was negatively correlated with the malignant grade of glioma;3 TIP30 and GDF-15,ZEB1 in glioma positive expression was negatively correlated,GDF-15 and ZEB1 in glioma positive expression was positively correlated,suggesting that TIP30 may play a role in the inhibition of GDF-15,ZEB1,And GDF15 may have a relationship with ZEB1.
