Objectives To observe the effects of Shizidaiping formula(SZDP)on glucose and lipid metabolism,related indexes of insulin signal transduction proteins PI3 K,P-PI3 K and GLUT4 expression in spontaneously diabetic GK rats and to discuss the mechanisns of the improvement on insulin resistance in skeletal muscle,glucose and lipid metabolism.Methods Selected 60 SPF male SD rats,10 rats were randomly selected as the normal control group(Control)continued to feed with full diet,the others as the model of type 2 diabetes was established by feeding with high-fat diet for 4 weeks combined with intraperitoneal injection of STZ after adaptive feeding 1 week,detected fasting bloodglucose(FBG)and postprandial two hours blood glucose(PG2h),FBG?7.0 mmol·L-1,PG2h?11.1 mmol·L-1 was considered succeed T2 DM model.Model rats were randomly divided into 5 groups: the model group,Shizidaiping Formula groups of low,middle and high,the Metformin group.rats of Shizidaiping Formula groups were given high,medium and low dose of intragastric administration for a period of 8 weeks,normal group and model group were given normal saline.During the test,Rats were observed daily general state;take the tail vein of a very small amount of blood,random blood glucose test strip to detect fasting blood glucose,postprandial blood glucose two hours and the oral glucose tolerance tests were performed and analyzed before and after the drug administration.At the end of the 8 week,aortic blood was collected for the measurement of serum total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL),low density lipoprotein(LDL).Superoxide Dismutase(SOD),Malondialdehyde(MDA)and the Cpeptide levels tested by radioimmunoassay(ELISA).The level of PI3 K,P-PI3 K and GLUT4 in skeletal muscle were examined by Western blotting after SZDP Intervention.The data were analyzed by SPSS 20 software.Results 1 Normal rats in good condition,shiny coat,active.model group rats grow weeks of age with varying degrees of polydipsia,polyuria,rough coat,spiritual malaise,slow response and other symptoms;With the advance of treatment time,the drug group compared with the model group have a significant improvement.2 Compared with the normal group,FBG,PG2 h and AUC were significantly increased in the model group after 8 weeks of Administration(P<0.05 or P<0.01).Compared with the model group,the treatment group was significantly lower(P<0.05 or P<0.01).3 Compared with the normal group,model group after treatment Hb Alc,GSP,C-peptide,HOMA-IR significantly increased(P<0.05 or P<0.01);compared with the model group,each group of four indexes were significantly decreased,the difference was statistically significant(P<0.05 or P<0.01);4 Compared with the normal group,after the end of treatment,model group TG,CHO,LDL were significantly higher than the normal group,HDL was significantly lower than that of normal group(P<0.01);compared with the model group,metformin group and SZDP group significantly decreased TG and CHO levels,elevated levels of HDL(P<0.01),The difference of the LDL level between treatment group and model group(P>0.05)is not significantly,but has a trend to decreased LDL.5 Compared with the normal group,model group SOD was lower than that of normal group,MDA higher than the normal group,the difference was statistically significant(P<0.05 or P<0.01);compared with the model group,metformin group,SZDP-H,SZDP-M,SZDP-L group can significantly increase the level of SOD,decrease the level of MDA(P<0.05 or P<0.01)6 The results of Western blotting showed that there was no significant difference in the expression of PI3 K protein in skeletal muscle(P>0.05),and the expression of P-PI3 K and GLUT4 in the two groups(SZDP-H,SZDP-M,SZDP-L)was significantly higher than that in the model group(P<0.05)Conclusions 1 Shizidaiping Formula can reduce fasting blood glucose and postprandial 2 hours blood glucose,improve glucose tolerance,and reduce insulin resistance index in type 2 diabetic rats.2 Shizidaiping Formula can regulate the level of blood lipid,improve lipid metabolism disorder in T2 DM rats.3 Shizidaiping Formula can regulate the glucose and lipid metabolism and improve insulin resistance(IR)in rats with T2 DM model,and its pharmacological mechanism may be related to antioxidant stress and the expression of PPI3 K and GLUT4 in skeletal muscle.
