Hepatocellular carcinoma(HCC)has been one of the cancers that exhibit high morbidity and mortality rate in China.Besides,it is also one of the tumors that display poor prognosis.Diagnosis,treatment and prognosis of HCC have always been an important study in the research of scientists.In recent years,scientists have paid more and more attention to the relationship between miRNA and tumor.The abnormal expression of miRNA is closely related to the occurrence and development of HCC.Therefore,further study of the effect of miRNA on HCC and its mechanism has great value for understanding the mechanism of occurrence and development of HCC and the development of therapeutic strategies.It has been reported a lot about the relationship between mi RNA and HCC,and studies have shown that mi R-1188 can inhibit the proliferation of Hepa1-6 while promote its apoptosis.However,the mechanism of mi R-1188 and HCC is still unclear.To explore the role of mi R-1188 in HCC,by means of RNA-seq and bioinformatics software analysis,we found that Fgfr1 and Slc9a8 may be the target genes of mi R-1188 in Hepa1-6.Furthermore,Slc9a8 which was the target gene of mir-1188 was determined by using the dual luciferase reporter assay.Mi R-1188 suppressed the expression of Slc9a8 at mRNA level and protein levels by qRT-PCR and Western blot analysis.Previous studies have shown that IL-4 can inhibit the proliferation and migration of HCC,and due to mi R-1188 overexpression or inhibition,the expression of IL-4 changed in Hepa1-6.However,overexpression of Slc9a8 inhibited the expression of IL-4 mRNA,which was contrary to the results of mi R-1188 overexpression.Next,Slc9a8-overexpressing Hepa1-6 cells were transfected with mi R-1188 mimics.Importantly,the expression of IL-4 m RNA revealed that Slc9a8 down-regulation partially inhibited the up-regulation of mi R-1188.It was proved that mi R-1188 increased the expression of IL-4 m RNA by targeting Slc9a8,which indicates that miR-1188 can inhibit the proliferation of Hepa1-6through the regulation of IL-4.Finally,cell transplant assay in vivo showed that mi R-1188 targets Slc9a8,and overexpression of mi R-1188 increased the expression of IL-4 mRNA,consistent with cell experiments in vitro.In summary,Slc9a8 acted as the target gene of mi R-1188,and mi R-1188 increased the expression of IL-4 mRNA by targeting Slc9a8 in Hepa1-6 and in vivo,so as to inhibit the proliferation of hepatoma cells.
