Photodynamic therapy(PDT) is a different from the traditional method of treatment of toumor.The basic principle is the injection of photosensitizer drugs in cancer patients,and the drug can be targeted to enrich the surrounding tumor tissue.With a certain wavelength of light conditions,photosensitizer drugs absorb light energy to excite and react with tissue oxygen of tumor surrounding to form the higher activity of singlet oxygen.Singlet oxygen ultimately induces tumor cell death and reaches therapeutic targets.However,the poor tumor targeting,easy aggregation between the molecules and other defects limit the application of photosensitizer in PDT.In order to solve these problems,we have carried out the study of axial and biotargeting tin porphyrins as photosensitizer.Firstly,the tetraphenylporphyrin was prepared by Adler method.The axial tetraphenyltin porphyrin was obtained by pyridine method,in which Sn Cl2?2H2O provided a metal ligand.And the obtained axial tetraphenyl sin porphyrin was grafted by malonic acid and acetylated glucosamine.Secondly,the obtained tetraphenylporphyrin was sulfonated to get tetrakis(4-sulfophenyl)porphyrin with concentrated H2SO4 as sulfonating agent.The metal-tin coordination porphyrin compound was obtained by methanol method.Then,(4-sulfonatophenyl)tin porphyrin-hyaluronic acid was obtained by grafting hyaluronic acid.The structures of these two kinds of axial porphyrins were characterized by 1H NMR,UV-Vis and FT-IR spectroscopy.In addition,the quantum yields of the singlet oxygen of the two axial porphyrins were determined using 1,3-diphenylisobenzofuran(DPBF)as the capture reagent singlet oxygen.The results showed that these two porphyrin photosensitizers could produce higher singlet oxygen,whose yields were 0.68 and 0.73,respectively,indicating that the axial coordination can effectively solve the intermolecular self-aggregation of the porphyrin molecules and ensure high singlet oxygen yield.Therefore,it has potential application value in biomedicine field.
