ObjectiveThe study is designed to detect CDK5 mRNA and SIRT1 mRNA expression of the peripheral blood mononuclear cells in the coronary heart disease patients and the level of NO,FBG,hs-CRP and lipid in their serum.We do the preliminary research and discussion on CDK5 and SIRT1 involved in the pathogenesis of coronary heart disease,as well as the correlation of between CDK5 and NO,glucose lipid metabolism,inflammation,and to explore CDK5 involved in the mechanism of endothelial aging.MethodsWe selected 83 hospitalized patients from January 2016 to September 2016 in the department of cardiology in the First Affiliated Hospital of Guang Zhou Medical University.48 patients diagnosed coronary heart disease were examined by coronary angiography as coronary heart disease group,and then according to the coronary artery lesions,divided CHD group into CHD single-double vessel lesions group 20 cases and CHD multi-vessel lesions group 28 cases,and another 35 patients with negative results of coronary angiography as the negative control group.We detected CDK5 and SIRT1 gene expression level of the peripheral blood mononuclear cells in three group,as well as the level of NO,FBG,hs-CRP and lipid in the serum.And statistical analysis of CDK5,SIRT1 and NO relationship between each other,as well as correlation of between CDK5 and FBG,hs-CRP and lipid.Results1.CDK5 mRNA expression level of coronary heart disease group(1.206±0.219)is higher than the negative control group(1.023±0.220)(P<0.05);While SIRT1 mRNA expression level of coronary heart disease group(0.580±0.396)is significantly lower than the negative control group(1.279±0.806)(P<0.05);Serum NO level in coronary heart disease group(92.881±17.884)is significantly lower than the negative control group(106.838±17.237)(P<0.05).2.Compared CDK5,SIRT1 mRNA expression level with CHD single double-vessel lesions group,CHD multi-vessel lesions group and the negative control group.CDK5 mRNA expression level of CHD single-double vessel lesions group(1.190±0.222)is higher than the negative control group(1.023±0.220)(P<0.05),and CDK5 mRNA expression level of CHD multi-vessel lesions group(1.218±0.221)is also higher than the negative control group(1.023±0.220)(P<0.05),but there are no statistical significance(P>0.05)between CHD multi-vessel lesions group and CHD single-double vessel lesions group.SIRT1 mRNA expression level of CHD single-double vessel lesions group(0.623±0.395)is significantly lower than the negative control group(1.279±0.806)(P<0.05),and SIRT1 mRNA expression level of CHD multi-vessel lesions group(0.550±0.401)is also significantly lower than the negative control group(1.279±0.806)(P<0.05),but there are no statistical significance(P>0.05)between CHD multi-vessel lesions group and CHD single-double vessel lesions group.3.The results of correlation analysis show that CDK5 mRNA expression level has a negative correlation with SIRT1 mRNA(r=-0.226,P<0.05)and NO(r=-0.296,P<0.05);SIRT1mRNA expression level is positively related with NO(r=0.672,P<0.05).4.Correlation analysis between CDK5 mRNA expression level and blood glucose,blood lipids,inflammatory biomarker,the results show that CDK5 mRNA expression level has a positive correlation with FBG(r=0.242,P<0.05),TC(r=0.321,P<0.05),TG(r=0.332,P<0.05),LDL-c(r=0.273,P<0.05),hs-CRP(r=0.233,P<0.05),and a negative correlation with HDL-c(r =-0.243,P<0.05).Conclusion1.CDK5 mRNA low level expression exists in human peripheral blood mononuclear cells.2.CDK5 mRNA expression level of the peripheral blood mononuclear cells in the coronary heart disease patients is higher than the negative control group;SIRT1 mRNA expression level in patients with coronary heart disease is apparently lower than negative control group.CDK5 mRNA is negatively correlated with SIRT1 mRNA,suggesting that CDK5 may play an important role in the development of atherosclerosis and coronary heart disease.3.NO level in patients with coronary heart disease is apparently lower than negative control group.CDK5 is negatively correlated with NO,and SIRT1 is positively correlated with NO,suggesting that CDK5,SIRT1 may have closely relationship with NO formation.High expression of CDK5 and lower level of SIRT1 and NO may be related to the formation and development of endothelial aging and atherosclerosis.4.CDK5 mRNA is positively correlated with FBG,TC,TG,LDL-c,hs-CRP,as well as negatively correlated with HDL-c,indicating that CDK5 may participate in glucolipid metabolism and inflammatory reaction process.
