Objective Polycystic ovary syndrome has become a hot topic in the study of female infertility.A lot of studies have shown that micro RNAs play an important role in its pathogenesis.Our reaserch want to explore whether PCOS is a hereditary disease and the role of mi R-133 b in the pathogenesis of polycystic ovary syndrome and its mechanism.Method(1)Letrozole was used to induce the PCOS rat model,and we inject the PCOS rat model with mi R-133 b agomir and mi R-133 b antagomir respectively to change the mi R-133 b level in the ovary.We explore the changes of peripheral blood hormone and ovarian tissue under PCOS and after intervention as well as the expression level of target gene TAGLN2.(2)The pregnancy rate of each group of rats was recorded and calculated.(3)The offspring rats were sacrificed 6 weeks later,and the expression of mi R-133 b and target gene TAGLN2 were detectioned.Results(1)Under the optical microscope,compared with the control group,in PCOS mother rat ovarian,there are almost cystic follicles,and granulosa cells are significantly diminished.We can see rare developmental follicles and corpus luteum in the ovary.The normal ovarian struction is destroyed.The follicles and corpus luteum were found in the ovary of the PM mother group.Most follicles were primary follicles,and the granulosa cells were neatly arranged.Even the cystic follicles disappeared and the ovarian structure become normal.In the rat ovarian of PI group,follicles were mostly cystic expansion and granulosa cell layer decreased.(2)The level of serum T in PCOS model group was significantly higher than that in the other three groups(P <0.01).The serum T level in PM group was significantly lower than that in PCOS group,but there was no significant difference compared with control group.The levels of LH and LH / FSH in the PCOS group were significantly higher than those in the control group,and the difference was not statistically significant(P> 0.05)compared with the control group.The level of serum E2 in PCOS group was lower than that in the other three groups.The level of serum INS in the PCOS group was higher than that in the other three groups(P <0.05).(3)Real-time PCR showed that the level of mi R-133 b in PCOS group was significantly lower than that in control group(P <0.01),and the level of mi R-133 b in PM group was significantly higher than that in PCOS group(P <0.01).The expression of TAGLN2 in the PCOS group was significantly lower than that in the control group(P <0.05),and the level of mi R-133 b in the PI group was significantly lower than that in the control group(P < 0.01).(4)Western blot showed that the expression of TAGLN2 in the ovary of PCOS group was significantly decreased(P <0.01)compared with the control group.The expression of TAGLN2 in the ovary of PM rats was higher than that of PCOS group,and there was no significant difference compared with the control group.The expression level of TAGLN2 in PI group was also significantly decreased(P <0.01).(5)The pregnancy rate of PCOS group was significantly lower than that of control group(33.33% vs 100%,P <0.01),but the pregnancy rate of PM group was higher than that of PCOS group(83.33 % Vs 100%,P> 0.05).The pregnancy rate of PI rats was significantly lower than that of the control group(33.33% vs 100%,P <0.01).(6)In the ovary of the control offspring group,the follicles in different developmental stages and corpus luteum were found,and the oocytes of the mature follicles were clear,the multilayered granulosa cell were arranged neatly and the histological morphology of the ovaries was normal.The PCOS offspring group can be seen within a number of expansion of cystic follicles,and rare or absent oocytes.The granulosa cell layer was decreased and rare developmental follicles at all levels as well as corpus luteum.The PM offspring group can see some different developmental follicles and corpus luteum.The granulosa cell layer were arranged neatly.(7)The serum FSH level in the PI offspring group was significantly higher than that in the control group(P <0.05).The levels of LH and LH / FSH in serum in PI offspring group were higher than those in control group,but the difference was not significant(P> 0.05).The serum levels of E2 in the PCOS group,PM group and PI group were significantly lower than those in the control group.The level of serum INS in the PI group was lower than that in the other three groups,the difference was not statistically significant(P> 0.05).(8)Real-time PCR showed that the level of mi R-133 b in the ovaries of PCOS offspring group was significantly higher than that of the control group(P <0.01).The mi R-133 b of PM and PI group had no significant difference with the control group(P> 0.05).The expression level of TAGLN2 in PCOS offspring group was not statistically different from that in control group(P> 0.05).(9)Western blot showed that there was no significant difference in the expression of TAGLN2 in the ovaries of PCOS offspring group compared with the control group(P>0.05).The expression of TAGLN2 in the ovary of PM offspring rats was significantly lower than that of the control group(P <0.01).The expression level of TAGLN2 in PI offspring group was also significantly decreased(P <0.01).Conclution(1)Letrozole-induced PCOS model in SD rats is feasible.The changes of ovarian pathology and serum sex hormone levels in model mice are the same as those in PCOS patients.(2)The level of mi R-133 b in ovarian of PCOS group was significantly decreased.(3)Mi R-133 b can target the expression of TAGLN2.Increasing mi R-133 b and up-regulation of TAGLN2 expression can improve the symptoms of PCOS rats,most importantly,can increase the pregnancy rate..(4)The expression of mi R-133 b in maternal PCOS rats may affect the expression of mi R-133 b and its clinical phenotype.
