Objective:To observe the effect of lentivirus-mediated stem cell leukemia on C-kit m RNA and protein expression in guinea pig diabetic cystopathy.To lay the foundation for further research on the clinical treatment of diabetic cystopathy.Methods:Made guinea pig diabetic cystopathy by streptozotocin,36 cases were randomly divided into 3 groups,blank control group(n=12),negative control group(n=12),experimental group(n=12).The blank control group was gaven 0.2ml PBS solution;the negative control group was gaven lentivirus without SCL gene +PBS liquid 0.2ml;experimental group was gaven 2X107 IU lentivirus 0.2ml.Respectively killed 3 guinea pigs of each group after transfection on day 2?7?14d and 28,detected the C-kit m RNA and protein expression levels in the bladder by real-time fluorescence quantitative reverse transcription polymerase chain reaction and blot Western.Results:90 guinea pigs were injected with STZ,4 died after injected,and 38 DCP models were screened out.The success rate of the model was 44.2%.The expression level of c-kit m RNA in experimental group increased on day 2 after lentivirus transfected,significantly increased on day 7 and reached to the peak,then decreased at day 14 and day 28,but its level was higher than the blank control group and negative control group at each time point(p<0.05).The level of c-kit m RNA of blank control group and negative control group had no significant difference at each time point.The expression of c-kit protein in experimental group increased on day 2 after perfusiong,and significantly increased at day 7,when reached to the peak,then decreased at day 14,also its level was higher than the blank control group and negative control group(p<0.05);and the level of c-kit protein in blank control group and negative control group had no significant difference at each time point.But,on day 28,there was no significant difference in the expression of c-kit protein of three groups(p>0.05),which indicated the promoting effect of SCL disappeared gradually.Conclusion: The level of c-kit m RNA and protein can be significantly up-regulated after lentivirus-mediated human SCL gene successfully transfeced into guinea pig DCP bladder,and the expression can be sustained,stable,long-term,which also provides a new way for treatment of DCP.
