Objective To Detect 5-hydroxymethyl cytosine(5-hmc)expression in melanoma,analyze the correlation between 5-hmc and invasion,metastasis and prognosis of melanoma.Methods Immunohistochemical SP method was used to detect the expression level of 5-hmc in 67 cases of melanoma and 20 cases of nevi.Univariate and multivariate analyses were performed using the Cox?s proportional hazards regression model.Results The expression rate of 5-hmc was significantly lower in melanoma than in pigmented nevus tissue specimens 40.30%(27/67)vs.75%(15/20),5-hmc expression was closely correlated with lymphatic metastasis,Clark grade,depth of invasion and prognosis of melanoma(P < 0.05),but was of no significant difference among patients of different age,gender or nationalities(P > 0.05).univariate regression of 67cases' clinical pathological features and molecular bio-makers results:lymph node metastasis,Clark grading and clinical staging?5-hmc expression level are relevant to prognosis(P<0.05);age?gender?national have no correlation with the prognosis of MM(P>0.05).Multivariate regression of 67cases' clinical pathological features and molecular bio-makers results:lymph node metastasis age[?= 1.14,HR(95%confidence interval)= 3.13(1.61-6.10),P<0.001]?No surgical resection[?=-1.6,HR(95% confidence interval)=0.20(0.10-0.40),P<0.001]?lower expression of 5-hmc[?=2.29,HR(95% confidence interval)=9.88(3.38-28.88),P<0.001]are independent prognosis factors of MM.Conclusion The expression of 5-hmc may be associated with lymph node metastasis in patients with melanoma.The prognosis of melanoma may be associated with clinical staging,treatment strategies and the expression of 5-hmc.Additionally,The expression of 5-hmc may serve as a new index for predicting prognosis of melanoma.
