Background:Ulcerative colitis(UC)is one nonspecific chronic inflammatory disease,one kind of inflammatory bowel disease,the etiology is not yet clear.Although some noninvasive,convenient biomarker have certain value in detecting UC,However,their sensitivitives and specificitives are limited.These markers can not replace the need for endoscopic evaluation.Because of economic factors and the risk of operation,endoscopy can not be used as a screening method for ulcerative colitis.Some studies have shown that the expression of serum IL-12 B was associated with the activity of ulcerative colitis,and serum IL-12 B may be an effective marker for predicting the activity and prognosis of ulcerative colitis.Objective: To determine the correlation of expression level of serum IL-12 B with activity and mucosal healing of UC,and to evaluate serum IL-12 B as a biomarker for the diagnosis of UC,the expression level of IL-12 B in serum of patients is measured and compared in different periods of UC.Method: The clinical data and results of colonoscopy of 80 patients with ulcerative colitis in Qingdao city hospital and 20 healthy controls were collected.Serum IL-12 B concentration was measured by enzyme linked immunosorbent assay(ELISA).Clinical activity of UC was assessed by Montreal classification of inflammatory bowel disease.Endoscopic activity of UC was assessed by Mayo endoscopic score.The clinical and endoscopic Serum IL-12 expression levels of UC patients were evaluated.P < 0.05 was found to be statistically significant.Results: The expression level of serum IL-12 B in patients with UC was significantly higher than that in control group(2.75pg/ml,1.42pg/ml,P<0.05).Clinical activity period was significantly higher than that of clinical active period(2.75pg/ml,1.42pg/ml,P<0.05).There were significences between the two simples.The active period of UC was higher than the remission period(1.88pg/ml,1.74pg/ml,P < 0.05).The difference was statistically significant.The IL-12 B levels in the course of clinical changes were tested by paired t test.Spearman correlation analysis was used to detect the expression level of IL-12 B and the severity of clinical and endoscopic diseases.Spearman correlation analysis was used to detect the correlation between IL-12 B expression and clinical and endoscopic disease severity.Both were positively correlated(r=0.90,r=0.95,P<0.05).Conclusion: Serum IL-12 B concentration was correlated with the disease activity and mucosal healing in patients with ulcerative colitis(P<0.05).Serum IL-12 B expression level increased with the severity of UC disease(P < 0.05).These results suggest that serum IL-12 B is a potential biomarker for disease activity and mucosal healing.
