Objective:To observe the protective effects of induced pluripotent stem cells(ADSCs)vitreous transplantation on retinal ganglion cells(RGCs)in the rat model of glaucoma.Methods: The rat model of glaucoma was performed by cauterization of 4 episcleral veins(EVC)of Long-Evans male rat eyes.ADSCs were isolated from rat bone marrow,amplified in vitro and tagged with quantum dot nanocrystals.All Animals were grouped as(1)model of glaucoma and ADSCs group receiving 5.105cells/6?LADSCs,(2)model of glaucoma and DMEM group receiving 6?L DMEM,(3)normal ADSCs group receiving5.105cells/6 ? L ADSCs and(4)normal DMEM group receiving 6 ? L DMEM.Vitreous injections were performed into the anterior chamber of 3 weeks-hypertensive eyes and IOP was monitored once a week for 3 weeks.At the end of experiment,The eyes were enucleated,and flat mounts of the retina from both eyes were prepared on a slide and evaluated by histological and immuuhistochemical analysis and reverse transcription-polymerase chain reaction(q RP-PCR)analysis.The labeled RGCs were counted by a computerized image analyzer,and the labeled RGC rate was used for statistical analysis.Results:performed one week later,IOP of model of glaucoma and ADSCs group and model of glaucoma and DMEM group receiving 6?l DMEM is higher on average10.9±2.1mm Hg than normal ADSCs group,and the trend can last to the whole process of the experiment(6 weeks).After the Injections,IOP of model of glaucoma and ADSCs group decrease more than model of glaucoma and DMEM group,and the difference was statistically significant(P<0.001).The damage changes of the retina appears in model of glaucoma and MSCs group and model of glaucoma and DMEM group in histological and immuuhistochemical analysis.The countation of RGCs in model of glaucoma and ADSCs group is more than it in model of glaucoma and DMEM group.The amount of BDNF m RNA of model of glaucoma and DMEM group is more than normal DMEM group,normal ADSCs group is more than normal DMEM group and model of glaucoma and MSCs group is more than others(*P<0.05;**P<0.01;***P<0.001).Conclusion:The vitreous injection of ADSCs can decrease the apoptosis of RGCs in the rat model of glaucoma.,and has the protective effect on RGCs.
