Objective: To study the effect of DIAPH3 on the migration and invasion of triple negative breast cancer(TNBC).Methods: The expression of DIAPH3 was detected in the MCF-10 A,SUM159,MCF-7 and MDA-MB-231 using the q PCR and western blot.Up-regulation the DIAPH3 expression,the migration and invasion of TNBC were detected using Transwell.Finally,study the molecular mechanisms about DIAPH3 influence the migration ability of TNBC.Results: The relative expression of DIAPH3 in the MCF-10 A,SUM159,MCF-7 and MDA-MB-231 were 1.00±0.000,0.47±0.068,0.64±0.039 and 0.17±0.008,respectively.Compare with the MCF-10 A,the expression of DIAPH3 in the breast cancer of UM159,MCF-7 and MDA-MB-231 were significant reduced(P<0.05).Compare with UM159,the expression of DIAPH3 in the TNBC of MCF-7 and MDA-MB-231 were significant reduced(P<0.05).Up-regulation the DIAPH3 expression,the migration number in the Cell group,NC group and DIAPH3 group were 305.5±6.36,329±16.97 and 221±24.04,respectively,and the invasion number in the Cell group,NC group and DIAPH3 group were 96±8.25,103±18.59 and 24.5±5.68,respectively.Compare with the Cell group and NC group,Up-regulation the DIAPH3 expression were significant inhibited the migration and invasion.Further,up-regulation the DIAPH3 expression were significant inhibited the expression of Rho A,MMP-2 and MMP-9.Conclusions: The expression of DIAPH3 was significant reduced in the breast cancer and the expression were forward reduced in the TNBC.DIAPH3 which up-regulated were influenced the migration ability of TNBC via inhibiting the expression of the Rho A,MMP-2 and MMP-9.
