Objective: The purposes of this study was to investigate the expression differences and correlations of PRL-3 and RhoC in the normal tissues,precancerous lesions and gastric cancer tissues during different pathological stages.We further analyzed the relationship between those two proteins and the gastric cancer invasion,metastasis and prognosis.Methods: This research collected 80 cases of gastric cancer patients after radical gastrectomy in our hospitals during the period of January 2011-April 2016 which including 60 cases gastric cancer during the period of January 2011 to October 2012,and 20 cases of patients after gastrointestinal surgery with gastric cancer during September 2015 to April 2016 in the hospital.In addition,50 cases of precancerous lesions diagnosed with high-grade intraepithelial neoplasia were collected in the digestive endoscopy room of our hospital.The gastric cancer adjacent normal tissues were set as controls Gastric and the case group were gastric cancer tissues and precancerous lesions.The mRNA level and corresponding protein levels of PRL-3 and RhoC in the precancerous lesion group,gastric cancer group and its adjacent group were detected by real-time quantitative PCR and immunohistochemistry assay respectively.Combining with the clinical pathology and follow-up data,we aimed to find the relationship between the expression of PRL-3 and RhoC and the prognosis of patients.Results:(1)RT-qPCR results showed that the relative expression level of PRL-3 in gastric cancer was(3.45 ± 1.21)significantly higher than that in the corresponding adjacent group(0.84 ± 0.12)and precancerous lesion group(1.42 ± 0.65)(student's t test,P <0.05).The expression level of RhoC in gastric cancer group(3.62 ± 1.35)was also significantly higher than that in adjacent tumor group(0.87 ± 0.18)and precancerous lesion group(1.51±0.83)(student's t test,P <0.05).(2)The positive expression rate of PRL-3 protein in gastric cancer group(75%)was higher than that in PRMC group(25%)and precancerous lesion group(58%)(P <0.05).The positive expression rate of RhoC protein in gastric cancer group was 77.5%,which was higher than that in adjacent normal group(21.25%)and precancerous lesion group(40%)(P <0.05).(3)The expression pattern of PRL-3 and RhoC in gastric cancer tissues was closely related to the degree of differentiation,depth of invasion,TNM staging and lymph node metastasis in gastric cancer patients(all P<0.05),and the expression of PRL-3 and RhoC was positively correlated(PRL-3:rs=0.726,P<0.05,RhoC: rs=0.740,P<0.05).4.Kaplan-Meier survival curve analysis and Log-rank test showed that the survival rate of PRL-3 and RhoC in negative group was significantly higher than that of positive group(PRL-3: P = 0.032;RhoC: P = 0.014).Conclusion:(1)The expression levels of PRL-3 and RhoC mRNA and protein were higher in gastric carcinoma,suggesting both of them were closely related to the invasion and metastasis of gastric cancer.the expression of PRL-3 and RhoC was positively correlated,which indicating that both of them played an important role in the development and progression of gastric cancer.(2)The mRNA and protein expression level of PRL-3 and RhoC was gradually increased during the process that from the normal gastric mucosal,precancerous lesions to the ultimate gastric cancer,so the detection of this two proteinscan help improve the diagnosis efficiency of early cancer.Both of which are expected to be markers of early diagnosis of gastric cancer.(3)The expression of PRL-3 and RhoC was closely related to the prognosis of patients with gastric cancer.The survival rate of PRL-3 and RhoC positive expression group was lower than that of negative expression group.Therefore,both of them could be used as risk factors to evaluate the prognosis of gastric cancer.
