Objective: To detect the relative expression of Toll-like receptor 4(TLR4)at m RNA level in complete blood cells of healthy subjects and patients with bone and joint tuberculosis;And to detect the expression of tumor necrosis factor-?(TNF-?)and Interleukin-6(IL-6)in the two groups,so as to preliminary study the expression and significance of TLR4 in complete blood cells,and its correlation with serum levels of TNF-? and IL-6 in patients with bone and joint tuberculosis.Methods:(1)The relative expression of TLR4 m RNA in complete blood cells of 113 healthy subjects and 81 patients with bone and joint tuberculosis was detected by real-time quantitative PCR technique.And the differential expression in two groups was compared.(2)The serum levels of TNF-? and IL-6 in two groups were detected by enzyme linked immunosorbent assay(ELISA).And the differential expression in two groups was compared.(3)In the observation group,the correlation between the relative expression of TLR4 m RNA and the serum levels of TNF-and IL-6 and the correlation between serum level of TNF-? and IL-6 were analyzed.Results:(1)The relative expression of TLR4 m RNA in the control group was 0.417±0.082,and the relative expression of TLR4 m RNA in the observation group was 0.735±0.161.The difference of relative expression of TLR4 m RNA between two groups was statistically significant(P<0.01).(2)The expression levels of TNF-? and IL-6 in the observation group were significantly higher than that in the control group.The difference of expression levels ofTNF-? and IL-6 between two groups was statistically significant(P<0.01).(3)In the observation group,there were no significant linear correlation between the relative expression of TLR4 m RNA and the serum levels of TNF-? and IL-6(r=0.178,P>0.05;r=0.043,P>0.05).And there was no significant linear correlation between the serum levels of TNF-? and IL-6(r=-0.042,P>0.05)Conclusion:(1)The relative expression of TLR4 m RNA in complete blood cells of patients with bone and joint tuberculosis was significantly higher than that in healthy subjects,suggesting that the high expression of TLR4 gene in the m RNA level may be closely related to the immune regulation of bone and joint tuberculosis.(2)The serum levels of TNF-? and IL-6 in patients with bone and joint tuberculosis were significantly higher than that in healthy subjects,suggesting that It may be involved in the occurrence and development of bone and joint tuberculosis.(3)There were no significant linear correlation between the expression of TLR4 m RNA in complete blood cells and serum levels of TNF-? and IL-6 in patients with bone and joint tuberculosis.
