Establishing the nucleic acid beacon of agar magnetic beads HBsAg diagnostic kit,which improves the early stage diagnosis rate of hepatitis B and contributes to treat it.In this paper,the magnetic beads as vehicle will combine with nucleic acid beacon aptamer and anti-HBsAg goat polyclonal antibody that forms beads-Polyclonal antibody-antigenmonoclonal antibody-beacon complexes to bulid a sandwich immune PCR detection technology.Using the automatic nucleic acid extraction technology developed the test kit.The immune-PCR results showed that the repetition rate is 97%,and the detection has a good linearity and significant when detection threshold is 105 ~ 108 copies and protein concentration of 10-3~10-6mg/mL.Finally draws the conclusion is automatic nucleic acids extraction technology and sandwich immune PCR detection technology can accurately detect target protein.It repeatability is good and sensitivity higher 103 than traditional ELSIA,which showed the sandwich immune PCR detection technology and diagnostic kits based on beacon nucleic acid of magnetic beads AGAR had built successfully.We termed the advantages of the detection kit with strong specificity,high sensitivity,good repeatability,Small error and high degree of mechanization for hepatitis B surface antigen detection.It is very important that the diagnosis and treatment of early stage hepatitis B virus(HBV)infection in clinic.
