Objectives Observe the expression of signaling molecules of HIF-1-VEGF pathway in intestinal tissue of rats with superior mesenteric venous thrombosis,and discuss the effect of this pathway in SMVT.Methods 1 64 male SD rats randomly divided into two groups,sham operation group(group Sham)and experimental group(group SMVT),32 rats in each group.2 Group Sham: opened the abdominal cavity,did not block the blood supply,rats were sacrificed after operated 8h,24 h,48h,72h;group SMVT: established the simple peripheral model of superior mesenteric venous thrombosis by ligation method,rats were sacrificed after modeled 8h,24 h,48h,72 h.3 Observe the changes of abdominal cavity and intestinal tract;HE staining detected pathological changes in intestinal tissue of rats;Real-time q PCR detected the expression of HIF-1? and VEGF m RNA in intestinal tissue of rats;Immunohistochemistry and Western blot detected the expression of HIF-1? and VEGF protein in intestinal tissue of rats.Results 1 The result of gross observation of abdominal cavity and intestine intestinal was shown,in group Sham,abdominal cavity and intestinal tract showed no obvious changes;in group SMVT,the abdominal cavity appeared different degrees and different character of fluid,the vein showed different degrees of anger state,the intestine tract showed different degrees of swelling and blood stasis,and all gradually reduced with time.2 The result of HE staining was shown,in group Sham,intestinal tissue showed no obvious pathological changes;in group SMVT,intestinal tissue showed different degrees of red blood cells stasis,and the degree of stasis gradually reduced with time.3 The result of Real-time q PCR was shown,the expression of HIF-1? and VEGF m RNA in rats of group SMVT increased significantly compared with group Sham at the same time(P<0.05),and both showed a decreasing trend.4 The results of Immunohistochemistry and Western blot were shown,the expression of HIF-1? and VEGF protein in rats of group SMVT increased significantly compared with group Sham at the same time(P<0.05),and all showed a trend of first rise and then decreased.Conclusions 1 After the onset of SMVT,the expression of HIF-1? was increased,and regulated the downstream target gene VEGF,started the angiogenesis and growth process,which had great significance to the establishment of collateral circulation in ischemic intestinal tract.2 After the onset of SMVT,the expression of VEGF was increased under the regulation of HIF-1?,promoted angiogenesis and growth,which had a role in promoting the establishment of collateral circulation in ischemic intestinal tissue.3 After the onset of SMVT,intestinal tissue ischemia hypoxia activated HIF-1-VEGF pathway,signal molecules HIF-1? and VEGF expression increased synchronously,promoted angiogenesis and growth,accelerated the establishment of collateral circulation,reduced blood stasis and microcirculation disorders,reduced the incidence of transmural intestinal infarction.
