Objectives In this study,the rat model of severe traumatic brain injury(TBI)was used to detect the changes of cold inducible RNA binding protein(CIRP)and Caspase-3 protein expression in the hippocampal CA3 region.The role of cold induced RNA binding protein(CIRP)and the therapeutic effect of mild hypothermia on brain injury in the treatment of nerve injury were discussed.Objective to investigate the protective mechanism of CIRP on brain injury after closed head injury,and to provide a theoretical basis for the clinical treatment of closed traumatic brain injury.Methods 1.The model of severe closed brain injury in rats was established by Marmarou et al.In 1994.2 288 Wistar rats were randomly divided into sham operation group,suture immediately after craniotomy(Sham),traumatic brain injury group(TBI)and hypothermia group(MH),the three group was divided into 6h,12 h,24h and 48 h four groups,each group of each 24 / time point.3 MH group: on the basis of the TBI group rats were kept in a constant low temperature experimental ice blanket,with the anus table to view and adjust the temperature of the blanket,so that its body temperature maintained at 32 degrees C,continued 48 h.4 The rats in general and anatomical observation,observed the changes of morphology and behavior changes of rats and brain tissue injury in different time points after hyperemia;using immunohistochemical method to observe the change of cell apoptosis and CIRP in hippocampal CA3 region in rats and Caspase-3 by pathology after mild hypothermia treatment;using Westren blot method of CIRP and Caspase-3 protein expression in the hippocampal CA3 region in rats was observed by mild hypothermia treatment.5 Using computer real color image analysis system;data using SPSS statistical analysis software,the multiple samples were compared using one-way ANOVA and the mean values were compared with t test.The results of Sham group was set to 1,and the other groups were compared,the results were expressed as mean + standard deviation,the probability of significant difference was set to 0.05 that the difference was statistically significant.Results 1 through the observation found in the case of 32 degrees of stability.Traumatic brain injury of rats after subarachnoid hemorrhage were seen,punctate hemorrhage more common in brain parenchyma and in the dorsal brain stem;traumatic brain surface hyperemia is sufficient,all injured rats showed no focal brain contusion performance.According to the observation of gross anatomy,the trauma is severe closed craniocerebral injury,which is consistent with the requirements of this study.It was observed that with the prolongation of the injury time,the blood vessels of the brain surface were gradually reduced,and compared with the TBI group,the blood pressure of the two groups was slower than that of the MH group.2 immunohistochemistry showed that the Sham group in hippocampus of rats with occasional CIRP positive cells,pale cytoplasm staining,positive cells in neurons after injury,TBI group and MH group in cytoplasm were brown or dark brown coloration and deep,with damage time coloring gradually weak,statistics CIRP results showed that in the MH group compared with TBI group,more than 10%,9%,6%and 5%(P<0.05),Caspase-3 showed that MH group compared with TBI group,less than6%,8%,10% and 13%(P<0.05).3 Westren and blot results from the same group,CIRP in TBI group and MH group were in the injury of 6h reached the peak,with the injury time to extend its protein expression decreased,MH group than in TBI group at different time points were more than 24%,21%,18% and 14%(P<0.05);Caspase-3 and CIRP results quite the contrary,the expression of 6h in early stage of injury was the lowest,with the injury time
