Objectives To study the antitumor effect of adenovirus vector-mediated IL-28B(Adh IL-28B)on He La cell cervical cancer and to construct Balb / C nude mice He La cell carcinoma subcutaneously transplanted tumor model to study adenovirus vector-mediated Effects of IL-28B(Ad-h IL-28B)on He La cells with subcutaneous xenografts in cervical cancer and its antitumor effect.Methods(Ad-h IL-28 B and Ad-EGFP)were used to amplify cells and adenovirus vectors(Ad-h IL-28 B and Ad-EGFP).The titer of the virus(Ad-h IL-28 B and Ad-EGFP)was determined by HEK293 cells MTL assay was used to detect the proliferation of He La cells.The expression of TAST3,Bax and Bc L-2 protein was detected by Western blot.The cells were treated with He La.The tumor growth rate was measured by HE staining.The expression of Ap La cells in He La cells was detected by immunohistochemistry.The expression of apoptosis-related proteins was detected by Western blot.Western blot was used to detect the expression of apoptosis-related proteins in He La cells.Results 1 The titers of Ad-h IL-28 B and Ad-EGFP were 108 PFU / 100 u L,and the MOI of He La cells was 10.2 MTT results showed that Ad-h IL-28 B group was significantly higher than Ad-EGFP group and PBS group The absorbance was low and IL-28 B inhibited the proliferation of He La cells(F=7.707,P=0.021).3 We successfully established Balb / C nude mice He La cell cervical cancer subcutaneous xenograft model.4 Ad-h IL-28 B could significantly reduce the volume of transplanted tumor in Ad-h IL-28 B group compared with PBS and Ad-EGFP group(F=437.597,P<0.001).5 The results of HE staining showed that the number of tumor cells in the tumor tissue of Ad-h IL-28 B group was significantly lower than that of the control group,and no obvious tumor metastasis was found in the lung and liver tissues.6 Western blot analysis showed that Ad-h IL-28 B significantly increased Bax protein level(F=117.905,P <0.001),but decreased the level of Bc L-2 protein(F=18.398,P =0.001)and STAT3 protein(F=12.356,P =0.003).Conclusions 1 Ad-h IL-28 B can inhibit the proliferation of cervical cancer He La cells.2 Ad-h IL-28 B can inhibit significantly the growth of subcutaneous xenografts of He La cells in nude mice.3 Ad-h IL-28 B may regulate the level of Bc L-2,STAT3 and Bax protein through JAK-STAT pathway,thus affecting the proliferation of cervical cancer He La cells and the growth of transplanted tumor.
