Rheumatic heart disease is a heart valve injury caused by recurrent episodes of rheumatic fever.About 15 million people are infected with the disease each year,and 300,000 people die because of that.In recent years,many studies have shown that causes of rheumatic heart disease are associated with expression of several genes,with deep studies in the epidemiological and pathologic aspects.However,further exploration and studies at the molecule level are still needed.In this study,genes associated with rheumatic heart disease were identified by RNA-seq of samples,and the expression analysis was performed to lay the foundation for the follow-up study.Clinical samples come from patients' auricula dextra.Samples were divided into two groups as valvular disease and rheumatic heart disease.The rheumatic heart disease group was then divided into two subgroups by with or without thrombosis.Pathological features were observed by HE staining.According to the data analyzed by RNA-seq of all groups,UBAP2 L,SNORD9,TNNT3,TPT1,SCARNA21,ACTA1,TRO were screened tentatively as related genes.After Semi-quantitative RT-PCR and qRT-PCR,there was no significant difference among three groups in the expression of UBAP2 L,SNORD9,TNNT3,SCARNA21,ACTA1,TRO while the expression of TPT1 in the rheumatic heart disease group was significantly up-regulated by 37% than that in patients with valvular disease.Besides,in the rheumatic heart disease group,the expression of TPT1 in thrombosis subgroup was increased by 35.3% compared with the other subgroup,showing the association between TPT1 and rheumatic heart disease.Further clarifying the expression of gene TPT1,FISH results showed that TPT1 expressed in the cytoplasm.Fluorescence immunohistochemistry showed that TPT1 was a cytoplasmic protein and there is a significant up-regulation in the expression in the rheumatic heart disease group.In summary,this topic was based on RNA-seq data and obtained a gene,TPT1,associated with rheumatic heart disease.The expression pattern of TPT1 in rheumatic heart disease was preliminarily identified,which provided the experimental basis for further study of its relationship with rheumatic heart disease.
