As the dried root bark of ranunculaceae plants’ peony(Paeonia surulicosa Andr.),Cortex moutan,bitter,sheen and slightly to taste,cold,in the heart,liver and kidney meridian,has the effect of heat-cleaning,blood-cooling and activating blood circulation to remove blood stasis,which used to treat heat invasion of blood chamber,vomiting blood,fevering without sweat,the pain of fall,carbuncle swollen and score from toxins.And cortex moutan charcoal has effect of cooling blood to stop bleeding,which clinical used in the treatment of vomiting blood,metrorrhagia,and a variety of other bleeding caused by heatsyndrome.But there are rarely reported about the material base of the view which is called “a material basis of both sexes ” in Cortex Moudan before and after the processing,and its pharmacology action and mechanism research.Therefore,on the basis of our prophase research,this paper analysis the chemical constituents of the petroleum ether fraction of Cortex Moutan charcoal,and screens the known chemical composition with antihemorrhagic activity,in order to find out the material base of Cortex Moutan charcoal chemical composition and also provide scientific theory basis of clarifying its pharmacological mechanism of removing heat from blood to stop bleeding.This subject isolate the pure compound of the petroleum ether fraction of Cortex Moutan charcoal with silica gelcolumn chromatographic,preparative TLC(PTLC)and MCI gelcolumn chromatographic,sephadex LH-20 column chromatography and recrystallizing methods.Finally,28 compounds were separated and identified its construction with NMR,ESI-MS and other modern spectroscopy and chemical methods.There are Paeonol(1),3-(2-Furanyl)-1-(2-hydroxy-4-methoxyphenyl)-2-propen-1-one(2),3,6-Dimethyl-5-benzofuranol(3),1,2-Benzenedicarboxylic acid diethyl ester(4),α1,α2-Ddiethyl-1,2-Benzenedimethanol(5),β-Sitosterol(6),1,2-Benzenedicarboxylic acid dibutyl ester(7),Benzoic acid(8),Methyl tetradec-5-enoate(9),Heptadecanoic acid 2-hydroxypropyl(10),1,4-Dipropylcyclohexane(11),Tridecanoic acid methyl ester(12),Tetradecanoic acid 2,3-dihydroxypropyl(13),3-Decyloxy-1-propanol(14),1-(3,5-Dihydroxy-4-methylphenyl)-ethanone(15),Betulinic acid methyl ester(16),2,4-Dihydroxyacetophenone(17),1-(2,5-Dihydroxy-4-methoxyphenyl)-1-propanone(18),Betulinic acid(19),Oleanolic acid(20),4-methoxy-benzoic acid(21),1-(3-hydroxy-4-methoxyphenyl)-ethanone(22),2,5-dihydroxy-4-methylacetophenone(23).The compound 2 is a new natural products,,the compound 5,9,10,11,12,13 and 14 are isolated from Cortex Moutan charcoal for the first time.We also used moleculardocking to predict the antihemorrhagic active constituent in Cortex Moutan charcoal and find out 23 compounds which conform to Lipinski’s “Rule of Five” and have differences in antihemorrhagic activity,in order to study primarily on its hemostatic mechanism.
