| Background:Oral ulcer is the most common and frequently recurrent disease in oral mucosal diseases.Because of its etiology and pathogenesis is not yet fully clear,modern medicine has no exact treatment and specific drug.Traditional Chinese medicine focuses on dialectical treatment and treatment of root of disease,so traditional Chinese medicine has obvious advantages in treating oral ulcer.Kouyangkang has significant effects on the treatment of oral inflammation caused by yin deficiency and fire excess.Objective:In order to evaluate the effect of Kouyangkang on the oral ulcer with yin deficiency in rats,and explore its mechanism from the p38 MAPK and ERK1/2 signaling pathway,then providing experimental basis for the further research and development of Kouyangkang.Methods:1、20 SD rats were randomly divided into two groups,with 10 rats in each group.The model rats were given thyroid plate suspension for 21 days.The blank rats were given normal saline.On the 22 nd day,the model rats were anesthetized,then the filter paper with a diameter of about 5 mm soaked in the 56%NaOH solution was placed in the lower lip mouth cavity mucosa of the rats burning for 20 s,the blank rats were not treated.After 24 h,the formation of ulcers were observed and the levels of cAMP and cGMP and the activity of Na+-K+-ATPase were measured.2、72 SD rats were randomly divided into 6 groups,including blank group,model group,zhibaidihuangwan group and kouyangkang high,medium and low dose group,with 12 rats in each group.By establishing the oral ulcer rats model with yin deficiency,studying the effect of Kouyangkang on the model rats.The levels of cAMP,cGMP,TNF-α,IL-6,IL-10,EGF,TGF-β1,the activity of Na+-K+-ATPase,and the expression of p-p38 MAPK,p-ERK1/2 protein were measured.Results:1、Compared with the blank group,the oral mucosa of the model rats formed ulcer with a diameter of about 5mm,and the body weight decreased,the drinking water and body temperature increased,tongue redness and so on.The level of cAMP and Na+-K+-ATPase activity were significantly increased,cGMP was significantly decreased(P<0.01).Under the microscope,the oral epithelial cells were dissolved and necrotic,and a lot of inflammatory cells infiltrated under the mucosa,mostly lymphocytes and neutrophils.Visible vascular endothelial cell necrosis and cellulose exudation formed necrotic tissue.2、Compared with the blank group,the model rats formed ulcers and had similar symptoms of Yin deficiency.The levels of cAMP,TNF-α and IL-6,the activity of Na+-K+-ATPase and the expression of p-p38 MAPK,p-ERK1/2 protein were significantly increased.The levels of cGMP,IL-10,EGF and TGF-β1 were significantly decreased(P <0.05,P <0.01).Compared with the model group,the symptoms of yin deficiency were significantly improved in the treatment group,and the ulcer healing time was shortened.The levels of cAMP,TNF-α and IL-6,the activity of Na+-K+-ATPase and the expression of p-p38 MAPK,p-ERK1/2 protein were significantly decreased.The levels of cGMP,IL-10,EGF and TGF-β1 were significantly increased(P <0.05,P <0.01).Conclusions:1、This study successfully establishes oral ulcer model with yin deficiency in rats,the model is simple and feasible,easy to repeat.2、Kouyangkang has obvious therapeutic effect on oral ulcer with yin deficiency in rats,and its mechanism may be achieved by regulating metabolism,related cytokine levels and phosphorylation levels of p38 MAPK and ERK1/2 protein. |
