Object: Esophageal cancer is one of the most common digestive maliganacies in the world,and the squamous cell carcinoma(ESCC)is the main histological type.ORAOV1,i.e.oral cancer overexpressed 1,has been identified within chromosomal band 11q13,which is one of the most frequently amplified lesions in human cancers.The gene/protein was amplified and overexpressed in several human cancers,and is associated with the invasion and lymph node metastasis of tumor cells,it is considered as a novel candidate oncogene.This study aimed to investigate the relationship of ORAOV1?vascular endothelial growth factor(VEGF)and microvascular density(MVD)in ESCC,and to discuss the role of ORAOV1 on angiogenesis of ESCC.Methods: The expression of VEGFprotein and CD31 MVD were assessed by using immunohistochemistry staining in 121 normal esophageal tissues(>5cm away from the tumor edge),96 esophageal low-grade intraepithelial neoplasia(ELGIN),53 esophageal high-grade intraepithelial neoplasia(EHGIN)and 143 ESCC specimens.Combined with our early work,the relationship among ORAOV1,VEGF and MVDwas assessed in ESCC.Moreover,the relationship of ORAOV1 and VEGF were examined by polymerase chain reaction(PCR)in ESCC cells in vitro.Results:(1)The VEGF protein(p<0.001)or MVD(p<0.001)gradually increased in normal esophageal?ELGIN?EHGINand ESCCtissues,which was consistent with the ORAOV1 protein.(2)The VEGF protein(p=0.012)or MVD(p=0.03)gradually increased with the increase of ORAOV1 protein in ESCC,andthe MVD gradually increased with the increase of VEGF protein(p<0.001).(3)In ESCC tissues,the protein of ORAOV1 showed positive correlations with VEGF protein(r=0.281,p=0.002),and MVD(r=0.291,p=0.008).And the VEGF protein and MVDalso showed positive correlation(r=0.373,p<0.001).(4)The VEGF expression(p<0.001)orMVD count(p=0.013)was higher in tumors with lymph node metastasis than tumors without lymph node metastasis,and the VEGF expressionwas higherin ?/? tumors than?/?tumors(p<0.001).(5)The silence of ORAOV1 gene in ESCC cells could reduce the expression of VEGF mRNA.Conclusion: ORAOV1?VEGFand MVDshowed significantly positivecorrelations in ESCC,and ORAOV1 gene regulates VEGF in ESCC cell lines.It indicates that the overexpression of ORAOV1 in ESCC may enhance angiogenesis by upregulating VEGF,and then paly important roles in promoting the invasion or metastasis of cancer cells.
