Objective Detecting the expression in lung of calcium sensing receptor(Ca SR)and 11?--hydroxysteroid dehydrogenase type2(11?-HSD2)in newborn mouse model of persistent pulmonary hypertension and to investigate the effect of calcium sensing receptor(Ca SR)agonists and antagonists on the expression of Ca SR and 11?-HSD2 in neonatal mouse model of persistent pulmonary hypertension;To clarify their roles in the PPHN model of neonatal mouse.Methods Newborn SPF C57BL/6 mouse pups and their mothers were exposed to 12% of the oxygen concentration,they were randomly divided into hypoxia group(PPHN group)(n = 11),hypoxia + agonist group(n = 13),hypoxia + antagonist group(n = 13);the control group of mouse pups and their mothers(n = 10)exposed in the air.The agonist group and the antagoist group received Gdcl3 16mg/kg and NPS2390 1mg/kg by intraperitoneal injection once daily,and all the treatment in each group lasted for 14 days.Pathological examination and immunohistochemistry were performed to examine the changes of pulmonary blood vessels.The expression of Ca SR and 11?-HSD2 in the lung tissue of neonatal mouse were detected by Confocal laser technology,q-RT PCR and Western blot.Results 1)Compared with the control group,the blood vessel wall thickness(WT%)and the thickness of the right ventricle and the left ventricular wall thickness(RV/LV)in the experimental group were significantly higher than those in the control group,and the model was successful;2)The expression of Ca SR in PPHN group was significantly increased compared with the control group,the agonist group was increased more obviously,and the expression in antagoist group was decreased(all P<0.05).3)The expression of 11?-HSD2 in the pulmonary vascular in hypoxia 14 days were decreased compared with the control(P<0.01).Gdcl3 further reduced,but NPS2390 enchanced the expression of 11?-HSD2 in the pulmonary vascular.Conclusion The animal model of persistent pulmonaryhypertension in neonatal mice is an effective and reliable method.The activation of Ca SR is involved in the persistent pulmonary hypertension,which may play an important role in the neonatal mouse.
