Cordia dichotoma Forst.f.belonging to the family of boraginaceae is a mature fruit of the Cordia dichotoma,and it's a traditional medicine in Uygur.Cordia dichotoma mainly grows in Yunnan,Guangxi,Guangdong,Taiwan,Fujian,Xinjiang and other warm place in China,but abroad its mainly distributied in India,Vietnam,northeastern Australia,the Philippines and other countries.Because of its role of clear abnormal mucus,it is widely used in the treatment of cold,cough,bravery,poor stool,high blood pressure and so on in clinical.The research of the specific medicinal ingredients and pharmacological effects of Cordia dichotoma Forst.f.is deficient,a comprehensive study of the active ingredients of rapeseed can be used to lay the foundation for the development and utilization of Cordia dichotoma Forst.f.In this paper,the research is divided into the following 4 parts.1.Optimization of high-speed shear treatment for extraction of total polyphenols from Cordia dichotoma Forst.f.using Box-Behnken experimental design: Based on the high-speed shear dispersing emulsifier technique,the effects of treatment time,rotary speed,liquid-solid ratio and ethanol concentration were investgatied.A predictive polynomial quadratic model was set up by means of central composite design and used to optimize these treatment conditions by response surface methodology.The optimal HSDE treatment conditions were determined as 3min of treatment time,19000r/min rotary speed,25 ml/g of liquid-solid ratio and 40% of the ethanol concentration,the comtents of total polyphenols were 8.74 mg GAE/g.Based on the kinetic theory,the extraction kinetics equation of polyphenols extracted from high shear shear emulsification at different rotary speed was established.The extraction process was in accordance with the second order kinetic equation and the equation was (?) DPPH free radical scavenging efficiency of polyphenols from Cordia dichotoma Forst.f.was studied,the extract of Cordia dichotoma Forst.f.(1.0 mg/m L)prossesses strong antioxidant as the same of ascorbic acid(0.4 mg/m L),and the clearance rate was up to 92%.2.Optimization of high-speed shear treatment for extraction of polysaccharides from Cordia dichotoma Forst.f.using Box-Behnken experimental design: Based on the high-speed shear dispersing emulsifier technique,the effects of treatment time,rotary speed and liquid-solid ratio were investgatied.A predictive polynomial quadratic model was set up by means of central composite design and used to optimize these treatment conditions by response surface methodology.The optimal HSDE treatment conditions were determined as 3 min of treatment time,16000 r/min rotary speed and 50 m L/g of liquid-solid ratio,the comtents of polysaccharides were 111.43 mg/g.3.Research of chmiscal constituents of enthol extract from Cordia dichotoma Forst.f.: The ethanol extracts were extracted by petroleum ether,chloroform,ethyl acetate and n-butanol respectively.The compounds were subjected to repeated silica gel column chromatography,dextran gel column chromatography and recrystallization to obtain 11 compounds,4 compounds were identified by spectroscopic techniques:4-ethoxy-4-oxobutanoic acid,4-hydroxy-3-methoxybenzaldehyde,(S)-2-hydroxy-3-(4-hydroxyphenyl)propanoic acid,4-Hydroxybenzoic acid,all of the above compounds were isolated from the plant for the first time.4.Synthesis of indoles and its derivatives: We have established for the first time tetrazole-1-acetic acid as an ideal ligand for promoting Cu I-catalyzed domino reaction of ortho-haloanilines with ?-keto esters/?-diketone for 2,3-disubstituted indoles' synthesis without the protection of an inert gas,20 substrates were synthesized.A variety of ortho-iodo/bromo-anilines and ?-keto esters/?-diketone could be reacted well to produce the target products in high yields,the yield between 32%-92%.The process can be applied to the synthesis of natural products which containing such groups.
