The Characteristics Of D2-40 Expression In Normal And Abnormal Lymph Nodes

Objective To observe the expression characteristics of D2-40 in normal lymph nodes and abnormal lymph nodes?Methods Ninty-two different kinds of lymph node samples from Beijing Friendship Hospital were retrospectively collected and(HE)sections were reviewed to confirm the diagnoses.All lymph nodes were studied using immunohistochemical staining for D2-40.Results 1.D2-40 was positive in the subcapsular sinus of normal lymph nodes(49/54),but negative or weakly positive in lymphatic sinus(43/54).However,there were four lymph nodes,in which the lymphatic sinuses enlarged and surrounded by a large number of plasma cells.The lymphatic sinuses in these four cases were strongly positive for D2-40.2.There were some irregular tubules in the interfollicular and paracortex showing strong immunoreactivity for D2-40,that is,lymphatic labyrinth reported in literature.3.In contrast to inactive lymph nodes(40/54),the quantity and staining intensity of lymphatic labyrinth was more and stronger in reactive lymph nodes.The difference was statistically significant(P <0.05).4.In the lymph nodes with metastatic cancer(15/36),the remaining subcapsular sinuses were positive for D2-40,while the lymphatic sinuses were negative.Cancer cells were surrounded by lymphatic labyrinth,not destroying the structure in 3 lymphnodes.5.D2-40 was also positive in the subcapsular sinus and negative in lymphatic sinus of most lymph nodes with lymphoma(16/21).Most lymph nodes had lymphatic labyrinth(18/21),but the staining intensity and quantity were different in each cases.Conclusions 1.D2-40 is not only expressed in lymphatic vessels but also in lymphoid sinuses,especially in subcapsular sinus.2.There is a large number of tubules positive for D2-40 in the interfollicular and paracortex of lymph nodes,that is,lymphatic labyrinth.It is the first time to use D2-40 to stain lymphatic labyrinth.3.The number of lymphatic labyrinth is more in the proliferative lymph nodes,and the staining is stronger,compared to normal lymph nodes.4.Compared to traditional scanning electron microscopy,it is more practical,simple and effective to show lymphatic labyrinth by D2-40,which provides a new method to study the network of lymph nodes.5.It is helpful to study the basic structure of lymph nodes by staining D2-40 in lymphoid sinus and lymphatic labyrinth,and understand the law how diseases form in lymph node...
