Objective To analyze the expression of lysosome-associated protein transmembrane 4 beta(LAPTM4B)in bladder cancer tissues,And further focusing on expression of LAPTM4B-35 and the clinical,pathological and prognostic significances in bladder cancer.Methods A total of 102 patients with bladder cancer were enrolled in this study.10 specimens were freshly resected for immunoblotting and 10 fresh resection of normal bladder mucosa were used in this study.Immunoblotting: The expression of LAPTM4 B encoded proteins is detected in 10 patients with bladder cancers tissues and 10 normal bladder tissues,using LAPTM4B-N1-99-p Ab.Immunohistochemical Staining: LAPTM4B1-99-p Ab was used to detect the LAPTM4B-35 expression in patients with 120 bladder cancer tissues.In addition,the relations between LAPTM4B-35 expression and the clinical,pathological characteristics of patients,were evaluated in this article.Statistical method: Using SPSS19.0 for Windows software for data analysis The correlation between the expression of LAPTM4B-35 in bladder carcinoma and clinicopathological factors was analyzed by chi square test Survival analysis: Survival rates were estimated using the Kaplan-Meier method.Log-rank test and Cox regression analysis were used in univariate and multivariate analysis,respectly.Results Ten frozen tissue sections of bladder cancers and ten frozen tissue samples of normal bladder were analyzed by immunoblotting to detect the LAPTM4B-35 expression.Normal bladder displayed almost negative expression and high expression was detected in bladder cancers.LAPTM4B-35 expression was found to have associations with TNM stage and histopathological classification by immunoblotting.It was shown that LAPTM4B-35 expression was closely related to bladder carcinoma lymphatic metastasis,invasiveness and recurrence rate.Among 102 patients with LAPTM4B-35 overexpression,56 patients were LAPTM4B-35 low-expression and 46 patients were LAPTM4B-35 high-expression.Univariate statistical analysis showed that the differences of the overall survival(OS)rates and the progression-free survival(PFS)rates between patients with high LAPTM4 B expression and low LAPTM4 B expression were both statistically significant.Multivariate statistical analysis showed that LAPTM4 B over-expression is an independent factor in epithelial bladder carcinoma prognosis(OS: HR=6.439,95% CI: 2.670-15.531,P<.0001;DFS: HR= 16.043,95% CI: 5.574-76.361,P<.0001).Conclusion 1.LAPTM4B-35 is positively overexpressed in the most patients with bladder cancers.2.LAPTM4B-35 expression is associated with clinical and pathological characteristics of patients with bladder cancer.3.Overexpression of LAPTM4B-35 may be associated with tumor progression and poor prognosis in bladder cancer,and thus may serve as a new molecular marker to predict the prognosis of bladder patients.
