Objective: To study the expression of Notch2 and Notch3 in non-small cell lung cancer(NSCLC),and determine its association with clinically relevant parameters(including gender,age,type of organization,TNM stage,tumor differentiation,lymph node metastasis,primary tumor size,primary tumor status),in order to discuss clinical significance of its expression in non-small cell lung cancer,Searching for new molecular markers that can be used to estimate the survival of NSCLC patients,as well as new targets for NSCLC targeted therapies.Methods: Select cancer tissue specimens of postoperative pathology confirmed 60 patients with NSCLC,at the same time take 30 cases of normal lung tissues adjacent to carcinoma specimens in comparison.By making paraffin biopsies,regular HE dyes,and immunohistochemical staining,to observe the expression of Notch2 and Notch3 in NSCLC tissues and normal lung tissues adjacent to carcinoma by immunohistochemistry.Results: 1.Notch2 was mainly expressed in cytoplasmof lung cancer cells,Notch2 was significantly up-regulated in NSCLC tissues than that in normal lung tissues adjacent to carcinoma(P <0.05);The expression of Notch2 in non-small cell lung cancer had no obvious correlation with patients' gender,age,type of organization and primary tumor size(P > 0.05);Notch2 positive expression rate in stage ? and ? non-small cell llung cancer patients was significantly higher than that in stage?and ? patients(P < 0.05),poorly differentiated obviously higher than that of high differentiation in patients(P < 0.05),patients with lymph node metastasis significantly higher than patients with no lymph node metastasis(P < 0.05),patients with primary tumor T3 and T4 significantly higher than patients with primary tumor T1 and T2(P < 0.05);2.Notch3 was mainly also expressed in cytoplasm,Notch3 was significantly up-regulated in NSCLC tissues than that in normal lung tissues adjacent to carcinoma;Notch3 positive expression rate in stage ? and ?non-small cell llung cancer patients was significantly higher than that in stage?and ?patients,poorly differentiated obviously higher than that of high differentiation in patients,patients with lymph node metastasis significantly higher than patients with no lymph node metastasis,patients with primary tumor T3 and T4 significantly higher than patients withprimary tumor T1 and T2,all of these differences are statistically significant(P < 0.05);The expression of Notch3 in non-small cell lung cancer also had no obvious correlation with patients' gender,age,type of organization and primary tumor size(P > 0.05);3.Notch2 and Notch3 expression in NSCLC were positively correlated(P < 0.05).Conclusion: Up-regulated expression of Notch2 and Notch3 was revealed in NSCLC tissues,Notch2 and Notch3 expression was closely related to clinical stages,degree of tumor differentiation,primary tumor status,lymph node metastasis or not,they play a role in the occurrence and development of non-small cell lung cancer,in judging the prognosis of patients with NSCLC have certain reference and guidance significance.
