Purpose This study selects three kinds of common unhealthy behaviors such as smoking,drinking,lack of sleep as research behavior,and focuses on the correlation between teenage unhealthy habits and immunologic imbalance of Th17?Treg?TNF-??sIL-2R.This study may have impant guiding value for the healthy growth of teenagers.Methods 1.Current research method was adopted to randomly sample 384 students of HZ Domestic Professional College.Questionnaire was self-made to do anonymous survey so as to analyze the current state of unhealthy habits and acne in adolescents.Chi-square test and logistic regression analysis were adopted to analyze risk factors of acne in adolescents with unhealthy habits.2.60 students participating in physical examination from September 2014 to December 2014 from HZ Domestic Professional College were chosen as research objects,and were divided into 3 groups including 20 people in smoking group,20 people in group lack of sleep and 20 people in the control group.The levels of sIL-2R and TNF-? in serum was determined by Enzyme linked immunosorbent assay(ELISA)method.The ratio of Th17 and Treg cells in peripheral blood was determined by flow cytometry;TNF-?,Forkhead Box Protein P3(Foxp3),Retinoic Acid Receptor-Related Orphan Nuclear Receptor?t?t(ROR?t)mRNA levels in peripheral blood was determined by RT-qPCR.One-way ANOVA and Spearson correlation analysis were taken to explore the correlation among smoking,lack of sleep and sIL-2R?TNF-??Th17?Treg.3.Based on clinical diagnosis and Pillsbury classification,60 adolescent acne patients admitted to department of dermatology,Shanxian County Central Hospital from March 2015 to August 2015 were selected and divided into 4 groups,namely,Acne Level?,Acne Level ?,Acne Level ?,Acne Level ? with 15 people in each group.Also,15 people doing healthy physical examination were chosen as control group.The levels of sIL-2R and TNF-? in serum was determined by Enzyme linked immunosorbent assay(ELISA)method.The ratio of Th17 and Treg cells in peripheral blood was determined by flow cytometry.One-way ANOVA and Spearson correlation analysis were taken to explore the correlation among acne severity and sIL-2R?TNF-??Th17?Treg.Results 1.According to Chi-square test,smoking,lack of sleep,and drinking are all suspected factors of acne.Whereas only smoking and lack of sleep are suspected factors of acne based on dual logistic regression analysis.2.The levels of sIL-2R and TNF-? in smoking group and group lack of sleep were significantly higher than those in the control group,and the difference was statistically significant;the proportion of Th17 cells,Th17/Treg was significantly increased,the difference was statistically significant;the relative expression level of TNF-? and ROR?t mRNA was significantly increased,the difference was statistically significant.Smoking amount,time of lack of sleep was positively and significantly correlated with sIL-2R,TNF-?,Th17 cell ratio,and Th17/Treg.3.The levels of sIL-2R and TNF-a in the group of patients with acne were significantly increased compared with those in the control group;the proportion of Th17 cells,Th17/Treg was significantly increased,the difference between which was statistically significant.Acne severity was positively and significantly correlated with sIL-2R,TNF-?,Th17 cell ratio,and Th17/Treg.Conclusion In gene and molecular level,this study proves that the unhealty habits such as smoking?drinking?lack of sleep may lead to the increase of Th17 cells in peripheral blood,the imbalance of Th17/Treg ratio,the high expression of ROR?t?TNF-AmRNA and the elavation of sIL-2R?TNF-aexpression in peripheral blood.This study also proves that this immunological imbalance is more obvious than that in ordinary people.So,the data of this study demonstrate that the immunological imbalance which caused by smoking and lack of sleep may play an important role in the pathogenesis of acne lesions.This study may have impant guiding value for the cultivation of young students' healthy habits.
