?Objective?The goal of this paper is to cultivate mice with the specific knockout gene featured by peroxisome proliferator-activated receptor-?coactivator-1,PGC-1?,and to explore the effects that the conditional knockout gene featured by peroxisome proliferator-activated receptor-?coactivator-1,PGC-1? has on mice's behaviors.?Methods?The heterozygous mice,which were originally introduced from Jackson Lab of America,and are PGC-1?flox/+ with SPF level,are expected to be fed and bred into the homozygous mice with PGC-1?flox/flox.Then they are placed with the mice carrying Dlx5/6-Cre-IRES-EGFP which are expressed through Cre recombinase by specific knockout gene featured by peroxisome proliferator-activated receptor-?,and their offspring are conducted genotype identification thereafter.Consequently,three kinds of mice,respectively homozygote,heterozygote and wild-type,with peroxisome proliferator-activated receptor-? coactivator-1,PGC-1?,are produced.Four experiments,namely,elevated plus maze,vertical experiment,tail suspension experiment,and forced swimming experiment will be applied to these three types of mice.The software called Etho Vision will be used to collect the data of mice's behaviors,and Stat View to do the calculation and analysis.?Results?1.The researchers successfully cultivate the homozygous mice with the knockout of PGC-1?,heterozygous mice with knockout of PGC-1?and the wild-type mice.The first express Loxp and loxp Cre,the second Loxp and Cre,and the third either Loxp or Cre.2.The results of the first behavioral experiment:The vertical open field and the elevated plus maze experiments showed that the homozygous mice were significantly hyperactive and less anxiety than the wild type ones;the tail suspension and forced swimming tests exhibited that the homozygous mice show significantly anti-depressive performance than the wild type ones.3.The results of the second behavioral experiment: the vertical experiment,researchers found the movement distance of homozygote mice are much farther and faster than wild mice while there is no statistics meaning between the latter two.In the process of the elevated plus maze,the frequency of the open arm entry and movement distance of homozygous mice are higher than the latter two.Similarly,there is no statistics meaning between the latter two mice.In the tail suspension process,the homozygous and heterozygous mice struggle longer time than the wild.Meanwhile,there is no statistics meaning between the former two mice.The former two mice keep static for a shorter time than the latter one,and there is no statistics meaning between the former two mice.In the forced swimming experiment,the researchers receive the same result as that in the tail suspension one.?Conclusions?From the above results,it is known that conditionally knocking out the gene with peroxisome proliferator-activated receptor-? coactivator-1,PGC-1?,the mice expresses anxiety and depression.The ability of anti anxiety and depression in wild-type mice was stronger with age,but there was no change in the anti anxiety and depression like behavior of the heterozygote and homozygote.
