PARD6A(Partitioning defective 6 homolog alpha)gene,as an important molecular scaffold protein,is mainly focused on cell polarization and cell asymmetry division in current study.Although there are few articles show that this gene may be associated with breast cancer and lung cancer[1,2].There also have a large number of articles reported that the polarity protein may affected tumor migration and invasion,besides,this gene has not been researched in ovarian cancer,an in-depth study of this gene may defined a new target gene for ovarian cancer to initiate new targeted drugs.So this paper aimed at PARD6 A.Firstly,researching the correlation between PARD6 A and ovarian cancer,detecting the effection on ovarian cancer cell lines by specificity silencing or overexpression of PARD6 A.Then,further studying the relativity of PARD6 A protein and clinical ovarian cancer.On the basis of exploring the clinical relevance,further researching the mechanism of PARD6 A in ovarian cancer.Western blot technology detected that PARD6 A gene was highly expressed in serous ovarian cancer cell lines SKOV3 and A2780;IHC technology detected that PARD6 A protein highly expressed in serous and mucous ovarian cancer tissues(serous is the most common histological type of ovarian cancer)rather than normal ovarian tissue.Using wound healing experiment,transwell chamber method,adhersion experiment and IF technology,detected that silencing of PARD6 A could inhibit ovarian cancer cell migration and invasion,attenuate cell adhersion ability.Meanwhile,overexpression of PARD6 A promoted anchorage independent growth,cell migration and invasion.Through ovarian cancer lung metastasis model experiment,verified that PARD6 A gene was silenced by shRNA could inhibit the metastasis of ovarian cancer cell SKOV3 in mice.About the mechanism of PARD6 A inhibited ovarian cancer migration and invasion,the paper preliminary detected that PARD6 A may be involved in Cdc42 and RhoA mediated signaling pathways which was associated with cell migration and invasion.In summary,this paper aims at examining the role and mechanism of PARD6 A in ovarian cancer.We hope to find a new target gene of ovarian cancer in order to improve the treatment and prognosis of ovarian cancer and to prevent tumor deterioration and recurrence.
