Dental caries as a high prevalence of oral infectious diseases has already been paid attention by people.Bacteria factors is one of the important factors to increase the incidence and prevalence of dental caries.Studies have confirmed that S.mutans is one of the most main pathogenic of caries.Thus,to prevent caries occurrence and development fundamentally,our research team has been designed and developed an antimicrobial agents that is C16LL-37,and it can be targeted to S.mutans.The targeting area of C16LL-37 is 16 aminso acids(TFFRLFNRSFTQALGK)of competence stimulating peptide of S.mutans,and the killing regions is LL-37,which has a ?-helical structure Cathelicidins peptides,and it comes from human body.More than,the targeting area and the killing regions area are connected by the linker regions(-GGG-).C16LL-37 has distinct specificity for Streptococcus mutans,and has the stronger antibacterial force,the researchists have verified.On this basis,this study will further dynamics of C16LL-37 sterilization,antibacterial peptide after effect,antibacterial mechanism and its security,stability and other biological characteristics were studied.Methods: 1.To test the kinetics of sterilization and antibacterial peptide effect after testing through the techniques of plate colony count.2.Using the SEM to watch the changes of S.mutans,when treatment by C16LL-37 under 32?M.3.Enzyme linked immunosorbent assay was used to evaluate the hemolytic activity and stability of C16LL-37.Results: 1.When treated by C16LL-37 under 64?M about 60,120 and 150 mins,the livability of S.mutans was 0;The livability of S.mutans was less than 10%,when treated by C16LL-37 under 32?M about 60 mins,and the livability was still in a state of decline after 120 and 150mins;The survival rate of S.mutans was 12.63% afterC16LL-37 treated at 16?M for 60mins;The survival rate of S.mutans has proliferation trend after 120 mins.2.The livability of S.mutans was 3.46%,when treated under 64?M for 30 mins.After C16LL-37 treatment at 64?M,the changes of S.mutans was wacthed by SEM,and we found that the shape was presented irregular,and the surface was rough.The S.mutans furthermore became splitting.3.The haemolysis rate of C16LL-37(?64?M)was less than 0.33%.4.The study showed that there were no significant influence on the antibacterial activity of C16LL-37 under different conditions for instance,temperature,pH,salinity and trypsin at low concentration(P>0.05).Conclusion: C16LL-37 has strong antibacterial activity for S.mutans,and it has low toxicity,and high stability.These characteristics of C16LL-37 implies good perspective in caries research and clinical application.The antimicrobial mechanism of C16LL-37 is not lead to bacterial resistance.
