Objective:To investigate different osmotic pressure solution on the proliferation of Human embryonic skin cell,Human immortalizde epidermal cell,Human umbilical vein endothelial cell,Epidermal stem cells.Methods: Primary cultured ESC was identified by Immunofluorescence.Make five different concentration of osmotic pressure solution of DMEM.The concentration was G1(hypotonic group:230mmol/L),G2(hypotonic group:262mmol/L),G3(control group:293mmol/L),G4(hypertonic group:321mmol/L),G5(hypertonic group: 351mmol/L).The HESF,HIEC,HUVEC,ESC were incubated 1h,3h,6h,12 h,24h,48 h,72h with five different concentration of osmotic pressure solution of DMEM.The cell morphology were observed under the inverted microscope,the proliferation of the cells was detected by cck-8 test.Result: CCK-8 detection results showed that the HESF,both hypotonic group and hypertonic group inhibited cell proliferation(P<0.05);HIEC,G1,6h,24 h,48h,cell proliferation was inhibited(P<0.05),12 h,cell proliferation was accelerated(P<0.05);G2,1h,72 h cell proliferation was accelerated(P<0.05),48 h,cell proliferation was inhibited(P<0.05);G4,cell proliferation was inhibited(P<0.05);G5,inhibited cell proliferation(P<0.05);HUVEC,G1,1h,3h,12 h,24h,cell proliferation was inhibited(P<0.05);72h cell proliferation was accelerated(P<0.05);G2,cell proliferation was inhibited(P<0.05);G4,inhibited cell proliferation(P<0.05);G5,cell proliferation was inhibited(P<0.05).ESC:G1,6h,24 h,48h,cell proliferation was inhibited(P<0.05);G248h,72 h,cell proliferation was inhibited(P<0.05);G4,1h cell proliferation was accelerated(P<0.05);12h,24 h,48h,72 h,cell proliferation was inhibited(P<0.05);G5,1h,3h,6h,cell proliferation was accelerated(P<0.05),12 h,48h,72 h,cell proliferation was inhibited(P<0.05).Conclusion: The proliferation of HESF?HUVEC?HIEC?ESC was closely correlated with osmolarity of culture medium,meanwhile the proliferation of HIEC?HUVEC was influenced by osmotic cultured time.The effect of osmotic pressure on cell viability was confirmed by this study,which provided a theoretical basis for clinical work and tissue engineering.
