Aims: We found some down-reguated circRNAs from 14 paired gastric cancer tissues and adjacent tissues in our previous gene chip detection.Validating its expression level and finding the relationship between its expression level and clinical pathological features of gastric cancer,which can provide theoretical basis for diagnosis and treatment of gastric cancer.Methods: A total of 42 pairs of gastric cancer tissues and adjacent normal tissues were obtained from gastric cancer surgical specimens between March 2016 and March 2017 at the first hospital of lanzhou university,China.The hsacirc0001666 levels in 42 paired gastric cancer tissues and adjacent non-tumor were detected by quantitative reversetranscription-polymerase chain reaction?q RT-PCR?.Then,the association between the expression level of hsacirc0001666 and the clinicopathological features?gender,age,tumor size,clinicopathological staging,lymph node metastasis,etc.?of patients with gastriccancer was further analyzed.A receiver operating characteristic?ROC?curve was generated to evaluate the diagnostic value.Results:?1?Hsacirc0001666 was first found to be down-regulated in gastric cancer tissues?P <0.05?.?2?In 42 cases of gastric cancer patients,the relative expression of hsacirc0001666between gastric cancer tissues and adjacent tissues were 2 times is meaningful.The number of down-reguated cases of has-cir-0001666 were 19,the number of up-reguated cases were 4,and the number of meaningless cases were19.?3?Its expression levels were correlated with differentiation?P <0.05?,lymphaticmetastasis?P <0.05?,TNMstages?P <0.05?.?4?The expression level of hsacirc0001666 in the diagnosis of gastric cancer,the area under the ROC curve was0.668,P<0.05,the sensitivity was 0.571,the specificity was 0.686.Conclusion: The expression of circRNA hsacirc0001666 was down-regulated in gastric cancer tissues and its expression levels were correlated with differentiation,lymphatic metastasis,TNM stages.These results indicated that hsacirc0001666 may be involved in theccurrence and development process of gastric cancer.Furthermore,it may be a novel new biomarker for the diagnosis of gastric cancer.
