| Objective1.Study of lncRNA and mRNA expression differences between the tumor tissues and tissues adjacent to tumor of pancreatic neuroendocrine tumor,analyse differentially expressed transcripts involved in cellular functions by bioinformatics.2.Study the expression the potential crucial lncRNA in pancreatic neuroendocrine tumor tissue samples.3.Study one of the potential important lncRNAs about the role in cell function in Bon1 cell line.MethodsUsing high-throughput chip technology analysis 5 pairs of pancreatic neuroendocrine tumor tissue and tissues adjacent to tumor on the differences between lncRNA and mRNA expression.Relying on bioinformatics analysis,forecast molecular functions of these different transcription to guide the further research.Choose some differentially expressed significant and stable lncRNAs,using Quantitative real-time PCR to assess their stability differences in human tissue samples.Choose lncRNA with stable differences in tissue samples to explore its fuctions in the tumor cells in vitro.Results1.Obtain the difference of pancreatic neuroendocrine tumor lncRNA and mRNA expression spectrum data.Based on analysis of lnc RNA subclasses,mRNA GO and Pathway analysis and lncRNA-mRNA expression network,to predict lncRNA significantly differentially expressed in the molecular function in tumor cells.2.Chip analysis results are consistent with quantitative PCR.LncRNA-uc001 lva.4,lncRNA-ENST00000445606,lncRNA-uc001 uzl.3,lncRNA-ENST00000393515 differentially expressed in more tissue samples.3.Knocking down lncRNA-H19 by shRNA in Bon1 cells slow cell proliferation,abate clone forming ability and weaken cell migration ability.In addition,Erk,pAKT308,pAKT473 and pSTAT3 expression are cut down,implying that lncRNA-H19 have influence on Bon1 cells through Erk/STAT3 and AKT/STA3 signaling pathway.ConclusionsThere is a big difference of lncRNA and mRNA expression spectrum in pancreatic neuroendocrine tumor between tissue adjacent to tumor and tumor.These differences can be used as a potential biomarker for disease diagnosis,can provide a direction to the research of tumor formation development mechanism.The lncRNA-H19 in tissue samples expresses lower,but in Bon1 cells express higher.Knocking down its expression inhibited cell proliferation clone formation,migration and promoting apoptosis. |
