Objective: To investigate the target cells and the best dose of effective monomer of Astragalus mongholicus and Salvia miltiorrhiza(calycosin and tanshinone ?A)on renal prorection in hypertension.Methods: The human renal tubular epithelial cells(HK-2),human renal glomerular endothelial cells(Hrgec),and mouse podocytes were cultured in vitro and induced by angiotensin ?(Ang ?)to build hypertensive injury cells.Calycosin and tanshinone ?A were divided into 0.4,2,10,50 mg/L dose groups,and respectively designed interactive experiments to get 16 dose combination.Valsartan(10-5mol/L)was used as the control drug,and the best target cells and the best concentration were detected by the cell viability(MTT),endothelial nitric oxide synthase(eNOS)and apoptosis rate.Results: After the intervention of different concentrations of Ang? with 24 h,there was no significant difference between the groups of podocytes;but the abnormal proliferation of HK-2 was increased and the apoptosis rate was increased;meanwhile the abnormal proliferation of Hrgec was increased and the apoptosis rate was increased.After the intervention of different concentrations of drugs with 24 h,there was no significant difference between the groups of podocytes;after the intervention of concentration ratio of tanshinone ?A 10mg/L+ calycosin 10mg/L or valsartan,the abnormal proliferation and apoptosis rate of Hrgec was decreased,the survival rate was significantly increased;after the intervention of concentration ratio of tanshinone ?A 0.4mg/L+ calycosin 10mg/L or valsartan,the abnormal proliferation and apoptosis rate of HK-2 was decreased,the survival rate was significantly increased.Conclusion:(1)The main target cells of renal injury in hypertension were renal tubular epithelial cell and glomerular endothelial cell.(2)The main target cells of the effective monomer of Astragalus mongholicus and Salvia miltiorrhiza(calycosin and tanshinone ?A)on renal injury were renal tubular epithelial cell and glomerular endothelial cell;the concentration of tanshinone ?A 10mg/L+ calycosin 10mg/L has obvious protective effect on glomerular endothelial cells,and the concentration of tanshinone ?A 0.4mg/L+ calycosin 10mg/L has obvious protective effect on renal tubular epithelial cells.
