Objective: 1.To explore the artesunate against lung cancer A549 and 95 D cells migration.2.To explore the mechanisms of Hu R in artesunate-induced inhibition of invasion and metastasis of lung cancer.Methods: 1.Combing the artesunate and Hu R related literature,learning the latest progress of Hu R in antitumor and the newest trend of Hu R related role in the tumor.2.Use from low to high concentrations of artesunate in non-small cell lung cancer A549,95 D cell line 0 h,24 h,48 h,with CCK 8 experiment to observe the effect on tumor cell proliferation.3.The lung cancer A549,95 D cell line were treated for normal medium and Contains 30ug/m L artesunate culture medium for 24 hours,With the scratch test and Transwell experimental observation of cell migration and invasion of change.4.The cells were treated with differentconcentrations of artesunate for 24 hours,useflow cytometry instrument to detect apoptosis cells.5.The cells were treated with concentration of 30μg/m L of artesunate for 24 hours,RNA was picked up,using RT-PCR to check Hu R、MMP-9 expression.6.Treated with artesunate for 24 hours,total protein was extracted and using BSA kits to determine the protein concentration,By Western blotting experimental detection A549,95 D cells Hu R,MMP-9 protein expression changes.7.Using SPSS16.0 for statistical analysis,P<0.05 means statistical significant differences.Results: 1.Through literature review and consolidation,found that artesunate has prominent role in anti tumor invasion and metastasis,but its related molecular mechanism is unclear.Hu R,as a kind of RNA binding protein can regulate m RNA stability,and recently found that Hu R to participate in a variety of transfer related molecular transcription regulation,and a variety of human tumor cells express Hu R.2.In this research involves the range of concentration and time,with the increase of given artesunate concentration and the extension of processing time,its inhibition on the lung cancer A549 and 95 D cells also gradually enhanced,and show that 95 d cells of artesunate sensitivity is higher than A549,low concentration of artesunate can significantly inhibit the 95 d cells proliferation.3.The experimental results show using 30μg/m L artesunate treat lung cancer A549 and 95 d cells can significantly inhibit both tumor cell migration to the scratch area and significant inhibiting A549,95 d cells invasion ability,also shows the role of artesunate on 95 d cells are more sensitive.4.Apoptosis experimental results show that artesunate has no effect on lung cancer cell apoptosis 5.The Hu R and MMP-9 m RNA expression of A549,95 d cells treated by artesunate decreased significantly.6.The Hu R and MMP-9 protein expression of A549,95 d cells treated by artesunate decreased significantly.Conclusions: 1.The experimental results show that artesunate can inhibit A549,95 D cells proliferation,migration,and invasionability,but not obvious effects on cell apoptosis.With low concentration of artesunate(30 mu g/m L)can significantly inhibit cell migration and invasion of the two groups.Prompt artesunate effect on lung cancer cell biology behavior may be done through different pathways.2.This study results suggest the role of artesunate on human lung cancer A549,95 D cells biological behavior may be related to its influence Hu R,MMP-9.
