[OBJECTIVE] To investigate detective method of the T790M mutation of drug resistance of epidermal growth factor receptor (EGFR) gene in patients with lung adenocarcinoma.[METHODS] This study used super amplification refractory mutation system(Super ARMS) and amplification refractory mutation system (ARMS) to investigate the T790M gene mutation in plasma ctDNA samples from 152 lung adenocarcinoma cases. Furthermore, ddPCR was used as the gold standard to validate all test results.Finally, we also evaluated the two detecting techniques' sensitivity, specificity and detective efficiency.[RESULT] We found' the T790M mutation frequency was 5.26% (8/152) by ARMS.It was lower than T790M mutation frequency identified as 11.18% (17/152,p=0.06)by Super ARMS. We found post-TKI treatment samples (14/33) were associated with higher T790M mutation frequency of 39.39% (p=0.03). Super ARMS'sensitivity was 100.00% which was significantly higher than ARMS (47.05%,p<0.01).[CONCLUSION] Super ARMS is a better technology for detecting T790M secondary mutation related to drug resistance in patients with lung adenocarcinoma,and it is recommended as a more effective detecting method..
