Objective:1.Building the model of bacteria-fungus polymicrobial biofilms in Titanium surface in vitro. And researching the feature of bacteria-fungus polymicrobial biofilms in Titanium surface.2. Researching the effect of bacteria-fungus polymicrobial biofilms in Titanium surface in Prostaglandin E2?PGE2?.Methods:1. Building a model of Candida albicans - Staphylococcus epidermidis polymicrobial biofilms in Titanium surface in vitro. Choosing the Titanium as the experimental material,while deviting the Candida albicans,Staphylococcus epidermidis and mixed group into three groups. The Titanium should be put into the microbial suspension for each of them to cultivate the microbial for 6h, 12h, 24h, 48h, 72h. Then take out the Titanium to research the feature of bacteria-fungus polymicrobial biofilms in Titanium surface by Crystal violet?CV? staining, XTT, Confocal laser scanning microscope?CLSIM?, Scanning Electron Microscope?SEM?.2. Putting PGE2 with the concentration of 50pg/ml, 100pg/ml, 200pg/ml,400pg/ml respectively into the Candida albicans-Staphylococcus epidermidis polymicrobial suspension. And put Titanium into the microbial suspension.After cultivating for 24h, take out the Titanium to research the effect of PGE2 in bacteria-fungus polymicrobial biofilms in Titanium surface in by Crystal violet staining, XTT, CLSM, SEM.Results:1. The results of CV staining and XTT suggest that mixed group has stronger ability to shape and grow the biofilm than single species groups.2. The longer time to cultivate, the more complicated for structure and thicker the biofilm will be. Staphylococcus epidermidis mixed growth with spore and hyphae of Candida albicans. Staphylococcus epidermidis adhere to Candida albicans form a very complicated net structure.3. PGE2 influence polymicrobial biofilm formation in a concentration depends way. The promotion effect of polymicrobial biofilm formation from strong to weak is 200pg/ml?100pg/ml?400pg/ml?50pg/ml.4. With PGE2, there is no distinct different for structure of polymicrobial biofilms. But the number of hyphae of Candida albicans and the number of Staphylococcus epidermidis which adhere to Candida albicans are increasing.Conclusions:1. The growth of polymicrobial biofilms in Titanium is faster and more complicated than single microbial biofilm.2. PGE2 promotes the formation of polymicrobial biofilms and the promotes effect depends on the concentration of PGE2.
